药用真菌桑黄的液体发酵培养基的配方优化筛选研究

【摘要】 为探索名贵药用真菌桑黄摇瓶发酵培养基较优配方,在探索到不同碳源、氮源、无机盐种类和不同浓度单因素试验结果的基础上,分别选取玉米粉、黄豆粉、氯化钾作三因素三水平的L9(34)正交摇瓶培养试验。摇瓶发酵置于摇瓶转速135 r/min,28℃温度下培养9 d,以桑黄菌丝体干质量为检测指标进行发酵培养基配方的优化筛选。初步获得桑黄摇瓶发酵培养基较优配方:玉米粉30 g/L,黄豆粉10 g/L,氯化钾2 g/L,KH2PO4 4 g/L、MgSO4 2 g/L、VB1 0.1 g/L,使桑黄菌在摇瓶发酵培养条件下其菌丝体干质量可达2.23 g/100 mL。初步获得桑黄摇瓶发酵培养基较优配方,为进一步的桑黄菌摇瓶发酵工艺研究奠定一定的基础。

【Abstract】 The current study aims to explore a favorable formula for the shaking flask fermentation culture of the valuable medicinal fungus Phellinus igniarius.Based on the single factor test results of different concentrations and different carbon sources,nitrogen sources,inorganic salts using corn flour,soybean flour and KCl the L9(34) orthogonal shaking-flask culture of three factors and three levels was carried out at the speed of 135 r/min,with a period of 9 days at the temperature of 28℃.The fermentation medium optimization was based on the mycelium dry weight for the detection formula.A favorable formula for the shaking flask fermentation culture was obtained preliminarily,which contained corn powder 30g/L,soybean powder10g/L,KCl 2g/L,KH2PO4 4g/L,MgSO4 2g/L,VB1 0.1g/L,and Phellinus igniarius mycelium dry weight was up to 2.23g/100mL in the shaking flask fermentation culture condition.A favorable formula for the shaking flask fermentation culture of Phellinus igniarius was obtained preliminarily,which laid a foundation for further research of Phellinus igniarius in shaking flask fermentation condition.
