央芪汤流浸膏敷贴剂对胃癌动物模型的凋亡作用研究

【摘要】 目的观察央芪汤流浸膏制剂对胃癌种植瘤瘤体凋亡作用的影响。方法将人胃癌SGC-7901细胞悬液皮下接种于裸鼠右侧背部皮下,当接种部位肿瘤长至最大径4-5mm时,裸鼠随机分为4组:央芪汤敷贴剂组;氟尿嘧啶组;联合用药组和生理盐水对照组。给药期间观察裸鼠的一般情况及移植瘤的生长状况,使用游标卡尺测量和记录肿瘤的最大径(a)最小径(b),绘制肿瘤体积的生长曲线;连续用药24 d,并继续观察4d后,颈椎脱位处死裸鼠,连同胞膜完整剥出肿瘤组织,电子天平称重;按体积计算药物的抑瘤率;采用Tunel法检测细胞凋亡。结果央芪汤流浸膏可以明显增加荷瘤裸鼠肿瘤细胞凋亡,凋亡指数与氟尿嘧啶组和对照组相比,有明显差异(P<0.01)。结论央芪汤流浸膏可以通过促进胃癌细胞的凋亡达到抗肿瘤的目的。

【Abstract】 Objective To investigate the effects of Yangqi decoction on human Gastric cancer SGC-7901 cells in nude mice.MethodSGC-7901 cell suspension were inject in nude mice at right back subcutaneous when the inoculation site of the tumor grew to 4-5mm,Mice were randomly divided into four groups,Yangqi Fluidextract group,Fluorouracil injection group,Yangqi Fluidextract + Fluorouracil injection group,Saline group.Observed the status of the general situation and tumor growth in nude mice,recorded the Maximum diameter(a) and trails(b) of the tumor.Draw the growth curve of tumor volume,after 24 days,Cervical dislocation mice,Stripping out the tumor tissue,and weight.Calculate the inhibition rate of the drug.TUNEL was applied to test situation of apoptosis.ResultYangqi Fluidextract can significantly increase the tumor-bearing nude mice tumor cell apoptosis.Conclusion Yangqi fluidextract can promote apoptosis of gastric cancer cells to anti-tumor.
