相关生长因子在肾纤维化中的动态变化及清利湿热药物对其影响的实验研究
Experimental Study about Dynamic Changes of Related Growth Factors in Renal Fibrosis and Influence of Traditional Chinese Medicine can Clear Heat-dampness away
【摘要】 目的动态观察转化生长因子-β1(TGF-β1)和肝细胞生长因子(HGF)在系膜增生性肾炎模型肾组织中的病理性互逆变化规律,研究清化湿热瘀血方药对病理性互逆变化的调节作用。方法复制系膜增生性肾炎大鼠模型,并以清化湿热瘀血方药(慢肾蛋白停)进行防治。分别于造模第2、4、8周末,检测各组大鼠肾组织TGF-β1、HGF,观察其动态互逆变化的规律及慢肾蛋白停的调节作用。结果模型组第2周以后TGF-β1开始升高,第4周以后快速升高,而慢肾蛋白停组第2周TGF-β1同样升高,但第4周以后则快速下降而呈相反变化;模型组第2周以后HGF持续降低,而慢肾蛋白停组HGF则持续升高亦呈相反变化,并均有显著差异(P<0.05)。结论系膜增生性肾炎肾纤维化过程与TGF-β1升高和HGF降低的病理性互逆动态表达有关,清化湿热瘀血方药可以调节这种病理性的动态互逆表达,延缓肾纤维化的发生。
【Abstract】 Objective To observ dynamic changes of related growth factors in renal fibrosis,and study Mechanism of Heat-dampness in chronic nephritis.Methods copy the models of MsPGN,and Chinese medicine were give to the manshen danbaiting group for preventive treatment.,at the end of the second week,the fourth week and the eighth week of immune model,Detect TGF-β1、HGF of the renal tissue.Observe ther changes.Results TGF-β1 of the model rats’ kidney was significantly increased which was tested at the end of the fourth week and the eighth week;Meanwhile HGFexpressi-on was significantly reduced.The results compared with the normal group were significantly different(P<0.05).After the preventive treatment by manshen danbaiting,The renal compared with the model group were significantly different(P<0.05);The expression of renal HGF was significantly higher than the model group,compared with the model group were significantly different(P<0.05).Conclusion MsPGN has the features of Heat-dampness syndrome.In model group,The renal tissue expression of TGF-β1 continues to rise,the levels of HGF were increased in the idney early acute injury.But with the persistent of disease and the over expression of TGF-β1,both levels were decreased gradually.Heat-dampness is the key facter of Renal fibrosis
