HPLC筛选千斤拔中黄酮类成分的水提工艺
Optimization of the extraction technology for the components in Moghania philippinensis by RP-HPLC
【摘要】 目的研究千斤拔有效成分的水提取最佳工艺条件。方法采用RP-HPLC法筛选,色谱柱为Diamonsil C18(250 mm×4.6 mm,5μm),流动相为甲醇-0.5%磷酸水(23∶77、10 min,43∶57、11~55 min),流速1.0 mL.min-1,柱温30℃,检测波长240 nm。结果千斤拔最佳水提取工艺的料液比为1∶16,80℃回流提取2 h,提取3次。结论所用工艺条件为千斤拔提取和进一步开发研究提供了参考。
【Abstract】 OBJECTIVE To optimize the extraction technology for Moghania philippinensis(Merr.et Rolfe)Li.METHODS The determination was carried out using a Diamonsil C18 column(250 mm×4.6 mm,5 μm),with methanol-0.5% phosphoric acid solution(23:77,10 min,43:57,11-55 minutes)as mobile phase at a flow rate of 1.0 mL·min-1.The column temperature was 30 ℃ and the detection wavelength was 240 nm.RESULTS The optimum technology of water extract was:solid-liquid ratio(1:16),80 ℃,2 h,extraction three times.CONCLUSION This experiment established the fundamental data base for further research of Moghania philippinensis(Merr.et Rolfe)Li.
