磷酸铝和鸡IL-18在新城疫F基因疫苗中的免疫佐剂作用(英文)
Immunological Adjuvant Function of Aluminium Phosphate and Chicken IL-18 in NDV F Gene Vaccine
【摘要】 [目的]研究磷酸铝、鸡IL-18分子佐剂pcDNA/chIL-18对新城疫F基因pcDNA/F的免疫佐剂作用。[方法]制备每份(0.2ml/只)含有磷酸铝佐剂(90μg)、pcDNA/F(200μg)、pcDNA/chIL-18(200μg)的疫苗,将实验鸡随机分为6组,每组12只,于7d龄分别肌注新城疫灭活疫苗、pcDNA/F+pcDNA/chIL-18+磷酸铝、pcDNA/F、pcDNA/F+pcDNA/chIL-18、pcDNA/F+磷酸铝、生理盐水;21d龄时以相同剂量进行2免。分别于免疫后0、7、14、21、28d采血,用ELISA检测其抗体水平,以淋巴细胞转化试验(MTT)方法检测T细胞转化率。35d龄时用30LD50的NDV强毒进行攻毒保护试验。[结果]pcDNA/F+磷酸铝+pcDNA/chIL-18免疫组的近期攻毒保护率达8/12,明显高于pcDNA/F组的保护率4/12和pcDNA/F+pcDNA/chIL-18组的保护率6/12;在ND抗体水平上,pcDNA/F+磷酸铝免疫组、pcDNA/F+pcDNA/chIL-18免疫组与pcDNA/F+pcDNA/chIL-18+磷酸铝免疫组无差异(P>0.05),均显著低于传统疫苗免疫组(P<0.05);pcDNA/F+pcDNA/chIL-18+磷酸铝免疫组的T细胞转化水平明显高于pcDNA/F免疫组(P<0.05),而pcDNA/F免疫组与传统疫苗免疫组无差异(P>0.05)。[结论]磷酸铝和pcDNA/chIL-18佐剂共同作用能明显提高NDVF基因疫苗的免疫作用。
【Abstract】 [Objective] This study aimed to investigate the immunological adjuvant function of aluminium phosphate and chicken IL-18 in NDV F gene vaccine.[Method] The vaccine(0.2 ml) containing aluminum phosphate adjuvant(90 μg),pcDNA/F(200 μg),and pcDNA/chIL-18(200 μg) was prepared.The 7 d old chickens to be tested were randomly divided into six groups(12 chickens in each group) and immunized through intramuscular injection with inactivated Newcastle disease vaccines,pcDNA/F+pcDNA/chIL-18+phosphate aluminum,pcDNA/F,pcDNA/F+pcDNA/chIL-18,pcDNA/F +aluminum phosphate,and physiological saline respectively;the secondary immunization was conducted with the same dose when the chickens were 21 d old.Their blood was sampled 0,7,14,21,28 d after first immunization.Antibody titer was detected with ELISA and T cell transformation rate was measured with MTT.Experimental chicken will be challenged with 30 LD 50 NDV virulence 28 d after first immunization.[Result] The survival rate of the chickens immunized with pcDNA/F +aluminium phosphate +pcDNA/chIL-18 achieved 8/12,higher than that of those immunized with pcDNA/F 4/12 and pcDNA/F+pcDNA/chIL-18(6/12).The NDV antibody titer of the chickens immunized with pcDNA/F+ aluminum phosphate,pcDNA/F+pcDNA/chIL-18 and pcDNA/F+pcDNA/chIL-18+aluminum phosphate is not different(P>0.05),but significantly lower than that of the chickens immunized with traditional vaccine(P <0.05).The T cell transformation rate of the chickens immunized with pcDNA/F +pcDNA/chIL-18 +aluminium phosphate was obviously higher than that of the chickens immunized with pcDNA/F(P<0.05).The T cell transformation rates of chickens immunized with pcDNA/F and the traditional vaccine showed no significant difference(P>0.05).[Conclusion] Combination of aluminium phosphate and pcDNA/chIL-18 can significantly enhance the immune effect of NDV F gene vaccine.
