参麦注射液对大鼠局灶性脑缺血的保护作用

【摘要】 目的观察参麦注射液对大鼠局灶性脑缺血损伤以及脑血流量的影响。方法采用大鼠大脑中动脉闭塞(middle cerebral artery occlusion,MACO)模型,观察参麦注射液对脑缺血大鼠神经功能、脑含水量、脑血流和脑组织超氧化物歧化酶(superoxide dismutase,SOD)、一氧化氮合酶(nitric oxide synthase,NOS)的活性及丙二醛(malondialde-hyde,MDA)、一氧化氮(nitric oxide,NO)含量的影响。结果参麦注射液能显著增加脑缺血组织的脑血流量(P<0.05或P<0.01);改善脑梗死范围和脑水肿(P<0.05或P<0.01),减轻脑组织海马区组织病理学改变;降低脑组织中NO、MDA含量和NOS活性(P<0.05或P<0.01);升高SOD的活性(P<0.05或P<0.01)。结论参麦注射液对大鼠局灶性脑缺血损伤具有良好治疗效果,可能与其抗氧化作用有关。

【Abstract】 Objective To investigate the effects of Shenmai injection on focal cerebral ischemia in rats.Methods Model of middle cerebral artery occlusion(MCAO) surgery was established in adult male Sprague-Dawley(SD) rats.After 24 hours of ischemia,the brain water content,infarction area of brain,cerebral blood flow and pathological changes in brain slices were observed.Superoxide dismutase(SOD) and nitric oxide synthase(NOS) activity,as well as malondialdehyde(MDA) and nitric oxide(NO) content in the homogenate of ischemic brain tissue were determined by spectrophotometric assay.Results Compared with ischemia group,the cerebral blood flow was elevated(P<0.05,P<0.01),the brain water content and the percentage of infarction area in the cerebral hemisphere were reduced(P<0.05,P<0.01),and the pathological changes in the hippocampus were ameliorated in Shenmai injection treatment groups.The content of NO and MDA,and NOS activity were decreased,but SOD activity in the ischemic brain tissue was increased in Shenmai injection group.Conclusion Shenmai injection has protective effects on focal cerebral ischemic injury,and the mechanism may relate to the antioxidant action via the decreased NOS activity and increased SOD activity.
