放线菌酮及二氮嗪在猪精液冷冻中的应用试验
An experiment with the application of cycloheximide and diazoxide for cryopreservation of boar semen
【摘要】 为了研究抗凋亡试剂在猪精液冷冻中的有效性,进一步优化猪精液冷冻稀释液配方,试验采用医学上常用的抗凋亡试剂放线菌酮(0,1.25,2.50,5.00μg/mL)和二氮嗪(0,0.25,0.50,1.00 mmol/L)加入改良后的冷冻稀释液中,对猪精液进行冷冻。结果表明:3种浓度的放线菌酮与对照组比较在猪精子稀释后活力、平衡后活力、冻后活力、精子路径速率(VAP)、直线运动速率(VSL)、轨迹(VCL)6个运动指标上没有显著差异(P>0.05)。3个组各指标的平均值都低于对照组。加入二氮嗪的3个试验组精子冻后活力都比对照组有所提高,但所检测的6个前述运动指标各组之间差异也均不显著(P>0.05)。说明在本试验条件下,放线菌酮和二氮嗪不能显著改善猪精子的冻后质量,2种试剂在猪精液冷冻保存中应用的适宜条件或必要性有待进一步研究。
【Abstract】 To validate the effect of anti-apoptotic reagents on the cryopreservation of boar semen,and to further optimize the formulations of diluents.Two kinds of anti-apoptotic reagents were added to the modified diluents,which included cycloheximide(at the concentration of 0,1.25,2.50,5.00 μg/mL) and diazoxide(at the concentration of 0,0.25,0.50,1.00 mmol/L),and then the routine cryopreservation procedure was conducted.The results showed that all the 6 parameters of spermatozoa in diluents supplemented with cycloheximide,which included the motility of extended boar semen,the motility of equilibrated and frozen-thawed spermatozoa,VAP,VSL and VCL of frozen-thawed spermatozoa),were not significantly different to those in the control(P>0.05);moreover,the average values of all above-mentioned parameters of treated spermatozoa were lower than those in the control;when the diluents were supplemented with diazoxide,the motility of frozen-thawed spermatozoa in the three groups was slightly increased,but the detected six motion parameters were not significantly different among the treatments(P>0.05).In conclusion,under the given experimental conditions cycloheximide and diazoxide don’t significantly improve the quality of frozen-thawed boar spermatozoa,which need to be further studied on the optimum conditions or necessity in the application of two drugs for the cryopreservation of boar semen.
