反相高效液相色谱法测定小鼠心肌中ATP、ADP和AMP含量及分析

【摘要】 建立同时测定小鼠心肌组织中ATP(三磷酸腺苷)、ADP(二磷酸腺苷)和AMP(一磷酸腺苷)含量的高效液相色谱方法。采用反相高效液相色谱模式,使用Sepax Bio-C18(250 mm×4.6 mm ID,5μm,200 )色谱柱,以60 mmoL.L-1磷酸二氢钾、60 mmoL.L-1磷酸氢二钾组成的缓冲液(pH=6.68)为流动相等比洗脱,流速:0.6 mL.min-1;紫外检测波长为259 nm,带宽为30 nm。三种目标组分在10 min内实现基线分离,平均加标回收率均在99%以上,线性范围为1~200 mg.L-1,检出限分别为20、20、25 ng。该法简便、准确,易于移植。

【Abstract】 To establish the simultaneous determination of the content of ATP,ADP and AMP in rat myocardial tissue,HPLC was adopted.It was determined by RP-HPLC with a Sepax Bio-C18(250 mm×4.6 mm ID,5 μm,200 )column,and UV detector at a wave length of 259 nm(BW 30 nm).Mobile phase was 60 mmol·L-1 KH2PO4 and 60 mmol·L-1 K2HPO4(pH=6.68) and flow rate was 0.6 mL·min-1.The separation was achieved within 10 min.All the calibration curves were linear in the mass concentration range of 1~200 mg·L-1.The average recoveries of the standard additions were over 97%,and the LOD were 20,20 and 25 ng for ATP,ADP and AMP respectively.This method was proved to be reliable and reproducible.
