高原低氧条件下大鼠骨髓细胞凋亡及沙利度胺干预效应研究
Apoptosis of Rat Bone Marrow Cells and Intervention Effects of Thalidomide under Condition of High Altitude Hypoxia
【摘要】 目的:观察高原低氧条件下大鼠骨髓细胞凋亡情况,并应用沙利度胺进行干预研究,探讨细胞凋亡在慢性高原病发生发展中的作用。方法:TUNEL方法检测高原低氧条件下及其在沙利度胺干预后的大鼠骨髓细胞凋亡指数,与对照组进行比较分析。结果:高原低氧条件下1天时大鼠骨髓细胞凋亡指数减低,低氧1天组为(1.34±0.65)%,对照组为(2.73±0.51)%(P<0.01);与低氧21天组比较,沙利度胺干预后细胞凋亡增加,低氧21天组为(2.65±0.98)%,干预组为(4.02±2.13)%(P<0.05)。结论:高原低氧条件下大鼠骨髓细胞凋亡减少,细胞凋亡可能是慢性高原病的重要发病机制之一。抗血管新生药物沙利度胺有促进高原低氧大鼠骨髓细胞凋亡的作用,可能成为慢性高原病治疗的新手段。
【Abstract】 Objective:To observe the apoptosis of rat bone marrow cells and intervention effect of thalidomide,an antiangiogenic drug,at high altitude to investigate effects of cell apoptosis on chronic mountain sickness(CMS).Methods:Apoptosis index of rat bone marrow cells and intervention of thalidomide was detected by TUNEL(terminal deoxynucleotidyl transferase-mediated dUTP nick-end-labeling) assay at different geographic altitude(2 260m control group vs 4 000m altitude group).The results were compared with apoptosis index of the control group.Results:As compared with the control group,the apoptotic index of rat bone marrow cells was decreased on day 1 of hypoxia(1.34%±0.65% vs 2.73%±0.51%,P<0.01).The apoptotic index was increased after treatment with thalidomide on 21 days of hypoxia(2.65%±0.98% vs 4.02%±2.13%,P<0.05).Conclusions:The apoptosis of rat bone marrow cells was reduced under condition of high altitude hypoxia,so cell apoptosis may be one of important pathogenesis of CMS.Thalidomide intervention can promote apoptosis of rat bone marrow cells at high altitude,which maybe a new therapy for treatment of chronic mountain disease.
