维生素D3联合地塞米松诱导大鼠破骨细胞的体外培养

【摘要】 目的:观察VitD3联合地塞米松对大鼠骨髓体外培养破骨细胞的影响,探索提高破骨细胞体外培养效果的有效途径。方法:实验分为两组,以维生素D3骨髓诱导法为对照组,以维生素D3联合地塞米松诱导SD大鼠骨髓细胞为实验组,通过Trap染色、甲苯胺蓝染色和扫描电镜观察鉴定破骨细胞,并对计数结果进行统计学分析。结果:实验组与对照组均获得Trap染色阳性及在体外有噬骨能力的破骨细胞,实验组破骨细胞计数(培养第7天)多于对照组,有统计学差异(P<0.05)。结论:与维生素D3单独诱导相比,维生素D3联合地塞米松体外诱导大鼠破骨细胞的效率更高。

【Abstract】 Objective: To establish a method for inducing rat bone marrow cells into osteoclasts in vitro,and to explore the effective way to improve the effect of culturing osteoclasts in vitro.Methods: Vitamin D3 bone marrow cell induction method served as control.Vitamin D3 combined with dexamethasone were used to induce Sprague-Dawley rat bone marrow cells into osteoclasts.The osteoclasts were identified by Trap staining,toluidine blue staining and scanning electron microscope(SEM) observation.The count results of osteoclasts were analyzed statistically.Results: Trap-positive osteoclasts which had the ability of bone absorption were obtained in the two groups.Statistical differences were found in the amounts of osteoclasts between the two groups(P<0.05).Conclusion: Compared with inducing by vitamin D3 alone,inducing the formation of rat osteoclasts by vitamin D3 combined with dexamethasone have better effect.
