不同温度及pH值对人结肠腺癌细胞模型的影响
Effects of Different Temperature and PH Value on Caco-2 Cell Model
【摘要】 目的:建立人结肠腺癌细胞系(Caco-2)细胞模型并对其药物研究的方法进行初步探讨。方法:通过测定跨细胞膜电阻(TEER)、荧光素通透量及碱性磷酸酶(AKP)评价Caco-2细胞模型,用MTT法考察HBSS缓冲液在不同pH值、不同温度及不同时间条件下对Caco-2细胞的毒性。结果:各项指标的测定值符合要求,表明本实验建立的Caco-2细胞模型的完整性、紧密性和通透性良好;进行药物Caco-2细胞吸收潜性研究时,药物作用4 h之内对Caco-2细胞模型没有影响。结论:建立的Caco-2细胞模型可用于药物吸收机制研究。
【Abstract】 Objective: To establish the Caco-2 cell model,and to explore the method for applying it in drug research.Methods: Transepithelial electrical resistance(TEER),the permeability of fluorescein and the expression of alkaline phosphatase(APK) were used to evaluate Caco-2 cell model.MTT method was used to detect the toxicity of HBSS buffer to Caco-2 cell on the condition of different pH,different temperature and cultivation time.Results: Test results of all the indexes met the requirements,which indicated that integrity,consistency and permeability of the established Caco-2 cell model were good.The model was not affected when Caco-2 cell was used in drug absorption study within 4 hours.Conclusions: The established Caco-2 cell model can be used to study the absorption mechanism of drugs.
