维生素E对P.y17XL感染BALB/c小鼠早期Th1应答的免疫调节作用

【摘要】 目的探讨维生素E对致死型约氏疟原虫感染BALB/c小鼠Th1免疫应答的调节效应。方法将6~8周、雌性BALB/c小鼠经腹腔接种1×106P.yl7XL寄生的红细胞,并随机分为实验组与感染对照组,实验组于感染前10d口服给予VE预处理。流式细胞术检测感染后第0、3、5天脾Tregs的百分含量;ELISA检测脾细胞培养上清中的IFN-γ、IL-10的水平;Griess反应检测脾细胞培养上清中一氧化氮的含量。结果与正常感染组比较:补充VE组Tregs增殖明显增多;补充VE组IFN-γ和NO分泌减少,而IL-10的分泌水平增加。结论在P.yl7XL感染早期,VE预处理可显著抑制Th1免疫应答效应,从而进一步加剧P.y17XL感染BALB/c的死亡进程。

【Abstract】 Objective To investigate the Th1 immunoregulatory effect of VitaminE(VE) in BALB/c mice infected with Plasmodium yoelii 17XL(P.y17XL).Methods Female 6-8-week-old BALB/c mice were randomLy divided into two groups.The mice in VE group were orally administered VE once a day for successive 10 days before P.y17XL infection.The level of IFN-γ,IL-10 and NO in the supernatant of splenocytes culture were detected by ELISA and Griess reaction,respectively.Results The percentage of Tregs was obviously higher in VE group.IFN-γ and NO was reduced in VE group,while IL-10 was increased.Conclusion Pretreatment with VE significantly suppressed Th1 immune response during the early stage of P.y17XL infection in BALB/c mice.Therefore,the course of infection was further aggravated.
