甲磺酸伊马替尼联合塞来昔布对胃肠间质瘤细胞增殖及凋亡的影响

【摘要】 目的观察甲磺酸伊马替尼和塞来昔布联合对胃肠间质瘤细胞增殖及凋亡的影响。方法甲磺酸伊马替尼及塞来昔布单独或联合处理胃肠间质瘤细胞株GIST-T1,MTT法检测药物对胃肠间质瘤细胞株的生长抑制,Western blot检测Bax及Bcl-2蛋白表达。结果甲磺酸伊马替尼与塞来昔布联合应用显著增强甲磺酸伊马替尼对胃肠间质瘤细胞增殖;和甲磺酸伊马替尼及塞来昔布单用相比,二者联用后胃肠间质瘤细胞Bax表达增高及Bcl-2表达降低。结论甲磺酸伊马替尼和塞来昔布联合应用显著抑制胃肠间质瘤细胞增殖,可能和促肿瘤细胞凋亡有关,其机制可能与Bax表达上调及Bcl-2表达下调有关。

【Abstract】 Objective To observe the effect of IM combined with celecoxib on proliferation and apoptosis in gastrointestinal stromal tumor cells.Methods GIST-T1 cells were treated with IM or celecoxib or in combination.MTT was used to determine the inhibition of GIST-T1 cells and Western blot was used to detect the expression of Bax and Bcl-2 in cancer cells.Results After treatment with IM combined with celecoxib,the inhibitory effect of proliferation was significantly stronger than those of IM or celecoxib alone(P<0.05).In comparison with IM or celecoxib alone,the level of Bax expression was higher and the level of Bcl-2 expression was lower in GIST-T1 cells treated by both two drugs.Conclusion The combination of Imatinib mesylate and celecoxib may inhibit the cell proliferation,which might be associated with the promotion of apoptosis.The mechanism probably correlates with the upregulation of Bax and the downregulation of Bcl-2 in gastrointestinal stromal tumor cells.
