B细胞淋巴瘤主要组织相容性复合物Ⅱ类分子表达水平研究

【摘要】 目的了解B细胞淋巴瘤淋巴组织和外周血淋巴细胞主要组织相容性复合物(MHC)Ⅱ类分子表达情况。方法采用实时荧光定量聚合酶链反应和Western印迹法检测B细胞淋巴瘤组织MHCⅡmRNA和蛋白表达,流式细胞仪检测外周血淋巴细胞MHCⅡ类分子荧光强度。结果 B细胞淋巴瘤组织MHCⅡmRNA和蛋白相对表达量分别为1.21±0.16和2.38±0.24,低于正常淋巴组织的7.68±0.48和17.31±1.08(P<0.05),外周血淋巴细胞MHCⅡ类分子平均荧光强度也由对照组的438.00±46.00下降为235.00±23.00(P<0.05)。结论 B细胞淋巴瘤组织及外周血淋巴细胞MHCⅡ类分子表达下降,外周血淋巴细胞MHCⅡ类分子检测可辅助早期诊断B细胞淋巴瘤。

【Abstract】 Objective To explore the expression level of major histocompatibility complex class Ⅱ(MHC Ⅱ molecule) in B-cell lymphoma tissue and blood lymphocytes.Methods B-cell lymphoma tissue MHC Ⅱ molecule mRNA and protein levels were detected by real time fluorescent quantitation polymerase chain reaction and Western blot respectively.Blood lymphocyte MHC Ⅱ molecular fluorescence intensity was detected by flow cytometry.Results B-cell lymphoma tissue MHC Ⅱ mRNA and protein relative expression levels were 1.21±0.16 and 2.38±0.24,lower than 7.68±0.48 and 17.31±1.08 of normal tissues respectively.Blood lymphocyte MHC Ⅱ molecular median fluorescence intensity decrease to 235.00±23.00 from 438.00±46.00 of normal control group(P<0.05).Conclusion MHC Ⅱ molecular mRNA and protein expression could decrease in B-cell lymphoma tissue and blood lymphocytes,detection of blood lymphocyte MHC Ⅱ molecular could assist early diagnosis of B-cell lymphoma.
