分子探针-酶联夹心杂交法测定水中微囊藻
Detection of Microcystis Aeruginosa with Molecular Probe/Enzymelinked Sandwich Hybridization Assay
【摘要】 微囊藻是分布最广的一类水华蓝藻,研究、建立微囊藻的快速检测方法具有十分重要的意义。针对藻蓝蛋白的基因间隔区域序列设计了一组可用于检测微囊藻种属的特异性探针,以实验室培养的微囊藻Microcystis aeruginosa为研究对象,建立了一种检测微囊藻的夹心杂交法。将该法用于华中科技大学校园内的景观湖———镜湖水样的检测,结果与显微镜观察和全细胞PCR的结果一致,表明试验建立的方法是有效的。
【Abstract】 Cyanobacteria of the genus Microcystis are common bloom-forming species in lakes and reservoirs worldwide,and the establishment of rapid determination method for Microcystis is of great importance.Based on sequence analysis of phycocyanin intergenic spacers(PC-IGS) from Microcystis strains,a set of genus-specific probes was designed,and a sandwich hybridization assay(SHA) was established to quantitatively detect Microcystis aeruginosa.The water samples from Jing Lake in Huazhong University of Science and Technology were analyzed with SHA,whole-cell PCR and microscopy.The results showed no distinct difference,indicating that the method was effective.
