超声波法提取贯众总黄酮及其不同极性部位的抗氧化作用
Ultrasonic Extraction of Total Flavonoids from Cyrtomium fortunei and Its Antioxidant Activities of Different Polarity
【摘要】 对超声波法提取贯众地下部分总黄酮的工艺条件进行优化,研究其不同极性部位抗氧化作用。采用紫外分光光度法测定提取物总黄酮含量,通过正交试验得到最佳提取工艺为:超声时间30m in,料液比1∶40(g/mL),乙醇浓度60%。采用有机溶剂萃取法将贯众地下部分提取物分氯仿相、乙酸乙酯相、水饱和正丁醇相和水相4个不同极性部位,以清除1,1二-苯基-2-三硝基苯肼自由基(DPPH.)、总还原力为评价指标,考察了不同极性部位提取物的的抗氧化作用。结果表明,4个不同极性部位提取物均有抗氧化作用,其中乙酸乙酯相的抗氧化作用最强。贯众根状茎具有较强的抗氧化活性,值得开发利用。
【Abstract】 The extraction process of total flavonoids from Cyrtomium fortunei rhizomes was optimized and the antioxidant activities of different polarity were studied.The total flavonoids of extraction was tested by ultraviolet spectrophotometry.The optimal process was obtained by orthogonal test experiments as follows: the extracting time of 30min,the ratio of raw materials to solution of 1∶40g/mL,and the ethanol concentration of 60%.4 extracts were obtained in different fractions by different polarities of solvents such as chloroform,ethyl acetate,water saturation normal butyl alcohol and water.According to the scavenging activity for 1,1-diphenyl-2-picrylhydrazyl radicals(DPPH·) and reducing activity,antioxidant activities of different polarity fractions were investigated.The results showed that four fractions all had different antioxidant effects,and the antioxidant activity of ethyl acetate fraction is the strongest.Extracts from Cyrtomiun fortunei show relatively strong antioxidant activities and are worth utilizing further.
