PCR法鉴定RorcγtGFP突变小鼠子代基因型
PCR-based genotype identification of filial generation in RorcγtGFP mutation mice
【摘要】 目的探讨PCR(聚合酶链式反应)法鉴定RorcγtGFP突变小鼠子代基因型的可行性。方法 Rorcγt(-/-)基因型雄性与Rorcγt(-/+)基因型雌性小鼠产生的6只新生小鼠,取尾部组织提取DNA,进行PCR扩增及琼脂糖凝胶电泳检测PCR产物,根据产物片段大小鉴定新生小鼠基因型。结果 3只小鼠PCR产物具有241bp片段,为Rorcγt(-/-)基因型;其余3只具有174bp及241bp片段,为Rorcγt(-/+)基因型。结论 PCR是检测RorcγtGFP突变品系小鼠基因型的简便、可靠方
【Abstract】 Objective To investigate the feasibility of PCR in the genotype identification of RorcγtGFP mutation mouse offspring. Methods The genome DNA was extracted from the tails of six newborn mice produced by a male Rorcγt(-/-) mouse and a female Rorcγt(-/+) mouse. Rorcγt gene fragments were amplified by PCR and then detected by agarose gel electrophoresis. Results Three mice had Rorcγt(-/-) genotype with 241 bp gene fragment, while three mice possessed Rorcγt (-/+) genotype with 174 bp and 241 bp gene fragments. Conclusion PCR-based method is simple and feasible for the genotype identification of RorcγtGFP mutation mouse offspring.
