猪源罗伊氏乳酸杆菌培养条件优化及其冻干保护剂筛选

【摘要】 从健康断奶仔猪中分离得到罗伊氏乳杆菌LR2,研究该菌株在不同pH值条件下的生长性能,确定罗伊氏乳杆菌最适培养pH值为5.5。采用恒定pH值分批培养方式高密度培养罗伊氏乳杆菌,最终发酵液中细胞密度达3.1×109CFU/mL。在菌悬液中添加海藻糖、脱脂奶粉等保护剂进行冷冻干燥,冻干菌粉活菌数为2.26×1011CFU/g,细胞存活率达81%。

【Abstract】 Lactobacillus reuteri LR2,isolated from healthy weaned piglets,was used as the original strain.The growth performance of Lactobacillus reuteri LR2 in different pH conditions was determined,and the result showed the maximum biomass was obtained at pH5.5.High cell density cultivation was carried out by a constant pH-controlled batch culture process,and a cell density reached 3.1×109 CFU/mL.The cell suspension mixed with trehalose,skim milk and other cryoprotectants during the freeze-drying process,the cell density of the freeze-drying production was 2.26×1011 CFU/g and the cell survival rate reached 81%.
