海洋微藻淀粉含量的测定
Detection of starch content in marine microalga
【摘要】 海洋微藻细胞内淀粉含量低,用常规碘显色法测定其淀粉含量时,细胞内残余的还原糖及色素干扰严重,所得结果误差大。为此,研究采用硝酸钙作为海洋微藻细胞内淀粉的提取剂,采用过氧化氢作氧化剂以有效消除残余还原糖及藻内原生色素对碘显色反应的干扰。结果表明,该方法的准确度和精确度符合分析要求,对富含淀粉藻种的筛选及优化培养条件的确定均有重要作用。
【Abstract】 The conventional iodine-colouration for assaying starch in marine microalga existed obvious deviation because of the low content of starch and the serious interference of reductive saccharides and pigment.So,calcium nitrate was used for extracting starch,and hydrogen peroxide was taken as oxidizing agent for eliminating the hindrance of eductive saccharides and pigment in microalga,the degree of accurancy and the degree of precision satisfied the analysis requirements.The results showed the experimental method was feasible and important for secreening the species of microalga abundant starch and determining the optimal culture conditions.
