用SRAP标记构建松树良种指纹图谱方法的研究

【摘要】 利用相关序列扩增多态性(SRAP)分子标记对三种松树各16个样本进行检测,旨在构建三个树种的指纹图谱,为松树品种鉴定提供依据。采用5个引物组合对样本进行了SRAP扩增,在湿加松、火加松和马尾松上分别检测到46、53和44个多态性位点,平均每对引物多态性位点分别为9.2、10.6和8.6个。根据扩增产物的电泳条带进行赋值,把SRAP图谱转换为由1和0组成的数字指纹图谱,仅用2个或3个引物组合就可以把同一树种的不同单株完全鉴别出来,为松树优良品系的鉴定提供了一种可行的方法。

【Abstract】 In order to construct DNA fingerprints for three species in Pinus,and further provide the basis for the variety identification,16 samples for each pine species were detected with Sequence-related Amplified Polymorphism(SRAP).Five pairs of SRAP primers were used,and 46,53 and 44 polymorphic loci were detected for P.elliottii var.elliottii×P.caribaea var.hondurensis,P.taeda×P.caribaea and P.massoniana,respectively,with 9.2,10.6 and 8.6 loci in average for each of primer in three species.The electrophoresis bands of amplified products were evaluated and SRAP band profiles were transformed to digital fingerprints consisting of 1 and 0.Different individual trees in different species were identified completely by using 2 or 3 pairs of primer only.This study provided a feasible method to identify pine improved varieties.
