低聚壳聚糖对辐射损伤小鼠细胞凋亡的影响
Effects of oligo-chitosan on apoptosis of radiated mice
【摘要】 从细胞凋亡角度探讨低聚壳聚糖提高辐射损伤动物存活率的机理。小鼠受到5 Gyγ射线照射后,检测骨髓淋巴细胞凋亡数、脾脏细胞凋亡相关基因和P53蛋白的变化。流式细胞仪检测细胞凋亡,RT-PCR技术检测凋亡基因,Western方法检测蛋白表达。与单纯照射组相比,低聚壳聚糖提高受照小鼠的存活率,降低骨髓淋巴细胞的凋亡细胞数量;下调凋亡相关基因Bax、caspas-3,上调Bc1-2,Bcl-2/Bax比值升高;抑制P53蛋白表达。低聚壳聚糖可能通过抑制P53蛋白表达,影响凋亡相关基因表达,从而抑制凋亡细胞的产生,有效提高受照小鼠存活率。
【Abstract】 In order to study the mechanism of oligo-chitosan boosting survival rate with irradiated mice,the apoptosis number of borrow leukomo-nocyte cell,apoptosis related gene and P53 protein expression of the mice after 5 Gy γ-rays irradiation was investigated respectively.The cell apoptosis was analyzed by flow cytometry.Apoptosis related gene was determined by Real Time-Polymerase Chain Reaction(RT-PCR) and Western blot assay was used to examine the expression of P53.Compared with the control group,the results show that oligo-chitosan can either in-crease the survival rate or decrease the number of apoptosis and apoptosis related gene of Bax caspas-3,while Bc1-2 and Bcl-2/Bax ratio increase and the expression of P53 is restrained.It can be deduced that oligo-chitosan will influ-ence the apoptosis related gene because of refraining the expression of P53 and leaded to shutdown the number of apoptosis,which then improve the survival rate of irradiated mice effectively.
