制备高效液相色谱法快速纯化粗榧中的山茶苷A
Quickly purification of camellianin A from Cephaltaxus sinensisby preparative HPLC
【摘要】 本实验是要研究粗榧中山茶苷A的分离方法,寻找一种高效液相色谱制备分离山茶苷A的快速简便方法,为粗榧药材及其黄酮类提取物的研究和应用奠定基础。实验采用的方法是用大孔吸附树脂柱色谱和硅胶柱色谱粗分,乙醇浓度梯度洗脱,得到山茶苷A粗品,用制备型高效液相色谱进一步分离纯化。结果表明,经制备色谱分离纯化的样品纯度达98%以上,光谱学初步鉴定为山茶苷A(芹菜素-5-O[α-L-吡喃鼠李糖基-(1→4)-β-D-6-O-乙酰基-吡喃葡萄糖苷])。本研究建立了利用高效液相色谱法,从山茶苷A粗品中分离制备高纯度山茶苷A的色谱方法,此方法简便易行。
【Abstract】 Objective:To establish a purification method of Camellianin A from Cephaltaxus sinensis.Methods:After extracted with ethanol,the extract of Cephaltaxus sinensis was isolated by macroporous adsorptive resins column chromatography and silica gel column chromatography, then purified by preparative HPLC.Results:The purity of product was greater than 98%by analytical HPLC,its structure was determined by chemical and spectroscopic methods as Camellianin A.Conclusion A purification method for preparation of Camellianin A from Cephaltaxus sinensis by preparative HPLC was established.The method is simple with lower cost,and higher sample purity.
