【摘要】 目的探讨胃癌组织中细胞凋亡情况,以及survivin、NF-κB在胃癌组织中表达,了解三者在胃癌发生、发展的作用和关系。方法共分为胃癌组织、癌旁组织、慢性胃炎组织、正常胃黏膜组织4组,分别采用TUNEL法检测组织中细胞凋亡情况,采取免疫组织化学法检测survivin、NF-κB表达情况。结果胃癌组织中survivin与NF-κB p65表达情况均较其他组高,而凋亡指数(AI)则最低(P<0.05)。survivin与NF-κB p65表达,凋亡指数与胃癌的临床分期、分化程度、淋巴结是否转移等密切相关。survivin与NF-κB p65在胃癌组织中表达情况呈正相关(r=0.289,P=0.01),survivin与NF-κB p65阳性表达的胃癌标本中凋亡指数均低于阴性表达的胃癌组织(P值均<0.01)。结论 survivin及NF-κB p65在胃癌组织中特异性高表达,对胃癌的发生、发展起促进作用,两者相互协同抑制肿瘤细胞凋亡可能是其中机制之一。联合检测survivin及NF-κB p65可能有助于评估胃癌的转移及预后。更多还原

【Abstract】 Objective To investigate the apoptosis and the expression of survivin,NF-κB in gastric carcinoma,and explore their relationships and roles in the pathogenesis of gastric carcinoma.Methods TUNEL method was used to detect apoptosis and immunohistochemic method was used to detect the expression of survivin,NF-κB in four group:gastric carcinoma group,adjacent cancer group,gastritis group,normal gastric mucosa group.Results The positive rate of survivin and NF-κB p65 in expression in gastric carcinoma group,was higher than other groups.While the apoptosis index of gastric carcinoma group occured smallest(P<0.05).The experssions of survivin,NF-κB p65 and apoptosis index were closely related to clinical TNM stage,differentiation and lymph node metastasis(P<0.05).It indicated that the expression of survivin in gastric carcinoma positively correlated with NF-κB p65(P<0.05).The apoptosis index of gastric carcinoma group which survivin,NF-κB p65 are positively expressed,were both significant lower than gastric carcinoma tissues that survivin,NF-κB p65 were negatively expressed.Conclusion Survivin and NF-κB p65 are expressed with a high specificity in gastric carcinoma tissues,and may play an important role in gastric carcinogenesis and development.It suggestes that synergistic inhibition of apoptosis may be one of the mechanisms.Combined detection of survivin and NF-κB p65 may be helpful for evaluation of the metastases and prognosis of gastric carcinoma.更多还原
