【摘要】 目的检测小肝癌患者血清蛋白及肝癌组织中微血管密度(MVD)、血管内皮生长因子(VEGF)的表达,探讨其对有微血管侵犯的小肝癌早期辅助诊断的意义。方法对58例小肝癌肝切除术患者的血清样本采用蛋白质飞行质谱技术检测蛋白表达谱;应用免疫组织化学方法检测患者肝癌组织的MVD及VEGF,分析微血管侵犯与蛋白表达谱及MVD、VEGF的相关性。结果 58例中,20例有微血管侵犯。蛋白质飞行质谱分析表明,微血管侵犯组在相对分子质量4 196,5 832的蛋白质峰明显升高,与无微血管侵犯组比较差别有统计学意义(P<0.05);肝癌组织MVD及VEGF检测表明,微血管侵犯组患者MVD计数及VEGF表达较无微血管侵犯组升高(P<0.05)。结论结合血清中蛋白质的差异性表达及肝癌组织MVD、VEGF检测,小肝癌微血管侵犯有望获得早期诊断。更多还原

【Abstract】 Objective To explore the values of three parameters: Serum protein,microvessel density(MVD) and vascular endothelial growth factor(VEGF) in early diagnosis of microvascular invasion of small hepatocellular carcinoma(SHCC).Methods 58 serum samples were collected from patients with SHCC,who underwent hepatectomy.Surface-enhanced laser desorption ionization time-of-flight mass spectrometry(SELDI-TOF-MS) was applied to analyze protein expression pattern,and immunohistochemistry was conducted to test MVD and VEGF expression on resected tissue from these patients.Statistical analysis was performed to assess the correlation between the three parameters and microvascular invasion in SHCC.Results 20 cases of 58 patients had microvascular invasion.As shown by SELDI-TOF-MS,samples with microvascular invasion had significantly increased in protein peaks with molecular weight 4 196 and 5 832;immunohistochemistry result showed that MVD and VEGF expression were also higher in samples with microvascular invasion.Compared with samples without microvascular invasion,the differences had significant statistical value.Conclusion The expression of serum protein,MVD and VEGF in SHCC paving the way for earlier diagnosis of Microvascular Invasion in SHCC.The detection of expression of serum protein,MVD and VEGF,paves the way for early diagnosis of microvascular invasion in SHCC.更多还原
