SELDI-TOF-MS技术用于结直肠癌诊断意义的分析
Diagnosis of Colorectal Cancer by SELDI-TOF-MS
【摘要】 目的探讨SELDI蛋白芯片技术在结直肠癌患者中的诊断价值。方法应用表面增强激光解吸电离飞行时间质谱仪和CM10芯片检测结直肠癌患者和构成比相似的对照组外周血清,筛选出结直肠癌特异蛋白峰并初步建立诊断模型。通过盲筛进一步验证诊断模型。同时收集上述结直肠癌患者根治术后7d的55例血清,观察手术前后有无差异蛋白峰。结果筛选出的特异蛋白峰质荷比为3 413,4 101,4 191,4 306,4 366,6 453,6 645,8 586,8 709,13 786,13 996,5 647,7 993和15 960Da,初步建立诊断模型。利用该模型对结直肠癌进行盲法验证,诊断的准确率为91.11%,灵敏度为88.89%,特异度为93.33%。根治术前后未发现差异蛋白峰。结论 SELDI技术可以筛选出结直肠癌患者血清特异蛋白并建立诊断分类树模型,有望成为结直肠癌早期诊断的辅助指标。
【Abstract】 Objective To explore SELDI-TOF-MS technology in diagnosis of colorectal cancer.Methods Surface enhanced laser desorption ionization time of flight mass spectrometry and CM10 chips technology was used to analyze serum specimens from 74 cases of colorectal cancer patients and 74 cases of healthy controls.Specific protein peaks were identified and a preliminary diagnostic pattern was established.This pattern was further validated by a blind test.Serum samples before and after operation from 55 colorectal cancer patients were also subjected to SELDI-TOF-MS analysis.Results The relative molecular weights of the specific protein peaks detected from colorectal cancer patient serum were 3 413,4 101,4 191,4 306,4 366,6 453,6 645,8 586,8 709,13 786,13 996,5 647,7 993,and 15 960 Da.A preliminary diagnostic pattern was established and was applied to screen colorectal cancer patients in a blinded study with an accurate rate of 91.11%,a sensitivity rate of 88.89%,and a specificity rate of 90.00% respectively.No significantly different protein peaks were detected in serum specimens from patients before and after operation.Conclusion SELDI-TOF MS technology can identify the specific protein peaks for colorectal cancer and a diagnostic pattern is established.It can be applied in early diagnosis of colorectal cancer.
