含水纳米材料冷冻电镜直接成像研究

【摘要】 对细菌、聚合物胶束、载药脂质体和脂质体立方晶等含水纳米材料进行冷冻电镜直接成像研究。结果表明,与负染色技术相比,文中利用冷冻电镜技术观察到了保存更完整的细菌膜结构和细菌的部分内部结构、精细的多层聚合物胶束结构、包裹在脂质体内的纳米药物以及脂质体立方晶的晶格结构。与负染色技术相比,冷冻电镜技术能避免染色假象、真空变形等缺陷,更真实地反映了有关纳米材料的原有结构特点。

【Abstract】 The bacteria,polymer micelles,liposome with nano-drugs and the cubosomes were studied by Cryo-transmission electron microscopy(cryo-TEM).By using cryo-TEM,the bacterial membranes were preserved much better,and the cytoplasmic structure of the bacteria could be even directly seen.The multilayer structures of polymer vesicles were kept well with no distortion.The nano-drugs inside the liposomes were clearly observed.The crystal patterns and structures of liposomes could be clearly observed.Compared with the normal TEM with negative-stain technique,the morphologies and structures of the nanomaterials were truly obtained by cryo-TEM without artifacts usually caused by staining and the vacuum damages.
