复合益生菌芽孢杆菌发酵培养基及条件的优化

【摘要】 为了获取活菌数高、抗性强、经济效益好的益生菌制剂,对复合益生菌芽孢杆菌制剂的培养基配方及各项发酵工艺参数进行了优化。选取枯草芽孢杆菌和凝结芽孢杆菌进行联合培养,并采用单因子试验和正交试验对其发酵培养基成分进行了优化,在此基础上对其发酵条件做了进一步优化。优化后最佳培养基成分为:废糖蜜初始pH值7.5,转速160 r/min,接种量8%,装液量40%。经优化后活菌数得到显著提高,达到6.62×1010CFU/mL。

【Abstract】 A study was performed to optimize the culture medium for compound probiotics of Bacillus and the various types of parameters of fermentation process,aiming to obtain probiotic preparations with large numbers of active bacteria,strong resistance and economic values.Bacillus subtilis and Bacillus coagulans were cultured together.The culture medium ingredients were optimized by single-factor test and orthogonal experiments,and the fermentation conditions were also optimized based on the above experiments.The ingredients of optimal culture medium were obtained as molasses 15 g/L,Indian meal 15 g/L,soybean powder 40 g/L,KH2PO4 1 g/L,K2HPO4 1 g/L,and calcium chloride 2 g/L.The best information were obtained as temperature 37 degrees C,initial pH 7.5,rotational speed 160 r/min,inoculation amount 8%,and liquid volume 40%.The number of active bacteria was significantly enhanced,reaching 6.62×1010 CFU/mL.
