【摘要】 目的从山茱萸果肉中提取山茱萸总皂苷(total glycosides from Cormus officinalis,TGCO),观察TGCO对白血病细胞HL-60的抑制作用。方法采用乙醚脱脂乙醇回流提取法,提取山茱萸中活性成分总皂苷,用紫外分光光度计检测山茱萸提取物中总皂苷含量。以白血病细胞HL-60为实验对象,用MTT法检测TGCO作用于白血病HL-60细胞后的抑瘤作用;在荧光倒置显微镜下观察TGCO作用于白血病HL-60细胞后的生长变化。结果经紫外检测TGCO提取物OD值为0.401,皂苷类成分浓度为0.0257 g/ml,含量为0.257 g/ml。MTT法检测TGCO在5～100μg/ml浓度范围内具有抑制白血病HL-60肿瘤细胞作用,荧光倒置显微镜下观察TGCO在25～50μg/ml浓度范围内能明显改变白血病细胞HL-60的生物学特性。结论 TGCO对HL-60癌细胞有明显的抑制作用。更多还原

【Abstract】 Objiect To investigate the inhibition of TGCO(total glycosides from Cormus officinalis) that extract from Cornus officinalis pulp to human acute myloid leukemia cell line HL - 60.Methods Extract TGCO by using 75%ethanol reflux,UV was used to determine the content of total glycosides.Leukemia cell line HL - 60 was used as in vitro model.HL - 60 was treated with TGCO at different concentrations,cell growth was analyzed with inverted fluorescence microscope and MTT assay.Results TGCO OD is 0.401,which detected by UV.The concentration of glycosiders is 0.0257 g/ml,the content is 0.257 g/ml.5 - 50μg/ml of TGCO could inhibit leukemia cell line HL - 60.Conclusion TGCO significantly inhibeted leukemia cell line HL - 60.更多还原
