NF-κB、Notch-1和Ki-67在结肠癌中的表达及其意义
The expression and clinical significance of NF-κB,Notch-1 and Ki-67 in colonic mucosa in patients with colon cancer
【摘要】 目的探索核因子κB(NF-κB)、Notch-1和Ki-67在结肠癌组织中的表达及其临床意义。方法收集泸州医学院附属医院病理科2008年12月至2010年1月存档结肠蜡块120例,其中结肠腺瘤20例;结肠癌90例,包括高分化腺癌45例,中分化腺癌33例,低分化腺癌12例;结肠正常黏膜组织10例。采用免疫组织化学的方法检测NF-κB、Notch-1和Ki-67的表达。结果 (1)在结肠正常黏膜-腺瘤-癌序列中,NF-κB的阳性率呈递增趋势,其在结肠癌组中的表达率高于正常黏膜组(P<0.05);Notch-1的阳性率呈递增趋势,其在结肠癌组中的表达率高于正常黏膜组(P<0.05);Ki-67的阳性率呈递增趋势,其在结肠癌组中的阳性表达率高于结肠腺瘤组及正常黏膜组(P<0.05)。(2)NF-κB的表达与结肠癌大小、不同侵袭深度,有、无淋巴结转移及分化程度有关(P<0.05);Notch-1的阳性表达率与结肠癌大小、分化程度显著相关(P<0.05);Ki-67的阳性表达率与结肠癌的组织分化程度及有、无淋巴结转移有关(P<0.05)。秩和检验显示NF-κB、Notch-1、Ki-67在结肠癌不同分化程度中的表达率不同,分化越低,阳性率越高(P<0.05)。(3)NF-κB、Notch-1、Ki-67在结肠癌组织中的表达呈正相关(P<0.01)。结论 NF-κB、Notch-1、Ki-67与结肠癌的发生、发展有关;Notch信号通路和NF-κB信号通路可能相互影响共同参与结肠癌的发生、发展。
【Abstract】 Objective The present experiment is to investigate the expression of NF-κB,Notch-1 and Ki-67 in colonic mucosa in patients with colon cancer,for the clinical diagnosis and usefull prognostic indicator.Methods The 120 cases of paraffin block from the department of pathology of the affiliated hospital of Luzhou medical college from December 2008 to January 2010,20 colon adenoma,90 colon cancer,with 45 well-differentiated,33 moderately differentiated and 12 poorly differentiated,10 cases of normal colon.NF-κB,Notch-1,Ki-67 expression was assessed by immunohistochemistry method.Results(1) We demonstrated that the positive rate of NF-κB in the sequence of normal colon mucosa-adenoma-carcinoma was gradually increased.The positive rate of NF-κB in colon cancer was higher than that of normal mucosa group(P<0.05).The positive rate of Notch-1 in the sequence of normal colon mucosa-adenoma-carcinoma was gradually increased.The positive rate of Notch-1 in colon cancer was higher than that of normal mucosa group(P<0.05).The positive rate of Ki-67 in the sequence of normal colon mucosa-adenoma-carcinoma was gradually increased.The positive rate of Ki-67 in colon cancer group was higher than that of colon adenoma group and normal colon mucosa group(P<0.05).(2) We founded that the positive rate of NF-κB in colon cancer concerned with tumor size,different depth of invasion,lymph node metastasis and extent of differentiation(P<0.05).The positive rate of Notch-1 in colon cancer concerned with tumor size and extent of differentiation(P<0.05).The positive rate of Ki-67 in colon cancer concerned with the differentiation degree and lymph node metastasis(P<0.05).The expression of NF-κB,Notch-1 and Ki-67 by rank sum test was distinguished at distinct differentiation degree in colon cancer.The lower differentiation,the higher positive rate was(P<0.05).(3) We also founded that the expression of NF-κB,Notch-1 and Ki-67 in colon cancer had positive correlation(P<0.01).Conclusion All above,our data imply that(1) the high expression of NF-κB,Notch-1 and Ki-67 in colon mucosa is related with the pathogenesis and development of colon cancer.(2) NF-κB signaling pathway and Notch signaling pathway may interact together in the pathogenesis and development of colon cancer.
