补肾方药对骨质疏松大鼠白细胞介素-1、肿瘤坏死因子-α及转化生长因子-β1的影响

【摘要】 目的:研究补肾方药对去卵巢骨质疏松雌鼠的治疗作用及其机制。方法:70只雌性SD大鼠分为5组:正常组、假手术组、模型组、雌激素组、补肾方药组。手术12周后开始给药,连续给药12周,放射免疫法(RIA)检测雌二醇(E2)、白介素-1(IL-1)、肿瘤坏死因子-α(TNF-α)含量,酶联免疫法(ELISA)检测转化生长因子-β1(TGF-β1)含量。结果:与模型组比较,补肾方药组血清E2显著升高(P<0.01),IL-1、TNF-α均显著减少(P<0.05,P<0.01),血清TGF-β1显著增加(P<0.01)。结论:补肾方药抗去卵巢雌鼠骨质疏松的作用机制可能与补肾方药可升高雌激素,下调IL-1和TNF-α表达,抑制骨吸收;同时上调TGF-β1表达,促进骨形成有关。

【Abstract】 Objective: To study the role of Chinese herbs of tonify kidney on treatment in postmenopausal osteoporosis female rats.Methods: 70 female SD rats were divided into 5 groups: normal,sham operation(Sham),osteoporosis model group,estradiol(E2) group,Chinese herbs of tonify kidney group.Administration of gastric infusion was executed 12 weeks after the the operation,12 weeks in total,the content of serum estradiol,interleukin-1(IL-1) and tumor necrosis factor-α(TNF-α)were tested by RIA.The content of transforming growth factor-β1(TGF-β1) were tested by ELISA.Results: Compared with the model group,the serum E2,TGF-β1 decreased obviously in Chinese herbs of tonify kidney group,and the difference from the treatment of E2 group was not significant.Conclusion: Chinese herbs of tonify kidney have significant improvement in the role of postmenopausal osteoporosis of female rats by upgrading the serum E2,and TGF-β1 to increase BMD of osteoporosis rats,which provided an experimental evidence of Chinese herbs of tonify kidney in clinic.
