清心开窍方有效成分对AD大鼠脑组织凋亡基因Bax、Bcl-2 mRNA及GFAP mRNA表达的影响

【摘要】 目的:探讨清心开窍方有效成分对AD大鼠学习记忆能力及脑组织细胞凋亡基因Bax、Bcl-2 mRNA及胶质纤维酸性蛋白(GFAP)mRNA表达的影响。方法:动物实验采用对照观察法。雄性SD大鼠56只,随机分为正常组、模型组、安理申组、清心开窍方组、皂苷组、多糖组和挥发油组,每组8只。采用双侧杏仁核注射Aβ25-35制造AD大鼠模型,造模后第2天起,分别灌胃给药安理申、清心开窍方水煎剂、皂苷、多糖、挥发油及双蒸水,2周后,观察大鼠水迷宫空间记忆能力,并通过RT-PCR研究脑组织Bax、Bcl-2 mRNA及GFAP mRNA的表达。结果:中药各组能明显改善AD大鼠学习记忆能力,大鼠大脑皮层、海马区Bax、GFAP mRNA表达水平降低,清心开窍方组、皂苷组皮质Bcl-2 mRNA表达水平升高,与模型组比较差异显著(P<0.05,P<0.01)。结论:清心开窍方能显著改善AD大鼠学习记忆的能力,可能是通过降低皮质、海马Bax mRNA及GFAP mRNA表达,升高Bcl-2 mRNA表达发挥作用。

【Abstract】 Objective:To study the effect of Qingxin Kaiqiao Formula and effective constituents on the expression of Bax,Bcl-2 mRNA and GFAP mRNA in brain of AD rats.Methods:Animal test used comparison method.Fifty-six male SD rats were randomly divided into seven groups: the normal group,the model group,the Aricept group,the Qingxin Kaiqiao Formula group,the saponin group,the polysaccharide group and the essential oils group,with 8 rats in each group.AD was induced by Aβ25-35 injected into the bilateral amygdale.The next day after molding,every group started to give medicines the Aricept,the Qingxin Kaiqiao Formula Decoction,saponin,polysaccharide,the essential oils and double distilled water,respectively.After 2 weeks,observed the spatial learning and memony ability,the expression of Bax,Bcl-2 mRNA and GFAP mRNA by RT-PCR(reverse transcription-PCR) in the brain of the animal.Results: Learning and memory ability of Qingxin Kaiqiao Formula groups was improved,the expression of Bax,Bcl-2 mRNA and GFAP mRNA decreased in the cortex and hippocampus,the expression of Bcl-2 mRNA of saponin group increased in the cortex,compared with model group,the difference was significant(P<0.05,P<0.01).Conclusion: Qingxin Kaiqiao formula may significantly improved in learning and memory ability of rats with AD by decreasing the expression of Bax,Bcl-2 mRNA and GFAP mRNA in the cortex and hippocampus.
