液相色谱-质谱技术测定大鼠血浆中L-NNA浓度

【摘要】 目的建立检测大鼠血浆中L-NNA浓度的液相色谱-质谱联用方法。方法采用Phenomenex Kromosil C18分析柱(4.6mm×250mm,5.0μm),以甲醇∶水溶液(10∶90)为流动相,电喷雾电离源,选择性离子监测质荷比(m/z)分别为:149.3(L-NNA)、218.2(D-苯甘氨酰胺),以D-苯甘氨酰胺为内标物,测定大鼠血浆中L-NNA的浓度。结果 L-NNA浓度在0.10~6.44mg/L范围内线性关系良好;最低检测限为24μg/L,最低定量限为80μg/L;血浆中L-NNA的回收率在85%以上,日内和日间的相对偏差(RSD)均<10%。结论本方法快速、准确、灵敏,可用于大鼠血浆中L-NNA浓度的测定。

【Abstract】 Objective To establish a LC-MS method for detecting plasma N-nitro-L-arginine(L-NNA)concentration in rat.Methods Phenomenex Kromosil C18 column(4.6 mm×250 mm,5.0 μm) was used;the mobil phase was methanol-aqueos solution(10∶90);the flow rate was 0.4 ml/min;electrospray ionization and selected ion mass spectral(m/z)were 149.3(L-NNA)and 218.2(D-phenylglycinamide);plasma samples of L-NNA were analysed using D-phenylglycinamide as internal standard.Results The linear range of L-NNA was at 0.1 to 6.44 mg/L.Minimun detection limit was 24 μg/L and the lowest quantitation limit for L-NNA was 80 μg/L.The mean recovery rate was 85%.The average RSD of within-day and between-day recovery(precisions)were below 10%.Conclusion The method in this study is fast,accurate, sensitive and suitable for measuring the plasma concentration of L-NNA in rat.
