溴氰菊酯对小鼠肝肾组织结构损害的形态学观察
MORPHOLOGY OF LIVER AND KIDNEY DAMAGE IN MICE INDUCED BY DELTAMETHRIN
【摘要】 目的研究溴氰菊酯(DM)对小鼠肝、肾组织形态学的影响及其机制。方法将30只小鼠随机分为对照组、DM 3.125mg/kg组、DM 6.250mg/kg组,每组10只,分别腹腔注射橄榄油、相应浓度的DM溶液3~5d,苏木精-伊红(HE)染色观察各组小鼠注射后第3、5天肝、肾组织的形态学变化,酶组织化学法检测琥珀酸脱氢酶(SDH)、单胺氧化酶(MAO)和碱性磷酸酶(ALP)在肝、肾组织中的表达情况。结果 HE染色可见注射后第3、5天DM染毒组小鼠肝脏肝索排列紊乱,肝血窦及中央静脉内淤血严重,炎细胞成团聚集,肾小管水肿,肾小球毛细血管扩张淤血。酶组化染色显示,注射后第3、5天DM各组肝、肾组织内SDH和MAO阳性表达较对照组增强;ALP阳性表达在肾脏较对照组增强,但在肝脏较对照组减弱,差异均有显著性(F=4.907~47.303,q=3.937~13.964,P<0.05)。结论低浓度DM通过改变各种酶活性影响肝、肾的代谢功能而导致肝、肾的损伤。
【Abstract】 Objective To study the influence of deltamethrin(DM) on morphology of liver and kidney in mice and its mechanism.Methods Thirty mice were evenly randomized to three groups: olive-oil control group,DM 3.125 mg/kg group and DM 6.250 mg/kg group.The mice were offered intraperitoneal injection of olive oil,and DM,respectively for 3-5 days.Tissues of liver and kidney were taken,prepared and HE stained for morphological study.The expressions of succinate dehydrogenase(SDH),monoamine oxidase(MAO) and alkaline phosphatase(ALP) in liver and kidney were measured by enzymohistochemistry.Results HE staining showed abnormal arrangement of hepatic cords in mice in DM groups,severe congestion in liver hepatic sinusoid and central veins of liver;and phlogocytes lumped,edema of renal tubules and telangiectasis and congestion of glomerularcapillary.Enzymohistochemical stain showed the positive expressions of SDH and MAO in liver and kidney in DM groups were higher than that in the control;and the expression of ALP in DM groups was higher than the control,but that in liver was lower,the differences between them were all significant(F=4.907-47.303,q=3.937-13.964,P<0.05).Conclusion Deltamethrin through increasing or decreasing various enzyme activity,affects metabolic functions of liver and kidney,leading to damage of the two organs in mice.
