发根农杆菌介导的橡胶树遗传转化体系的建立

【摘要】 [目的]建立新的橡胶树遗传转化体系,为利用基因工程技术研究橡胶树提供基础。[方法]利用发根农杆菌R1601、15834和1.2556 3个株系分别侵染橡胶树热研7-33-97的叶片和茎段,诱导毛状根,并利用PCR技术检测诱导得到的毛状根。[结果]在相同的条件下,仅橡胶树茎段在发根农杆菌R1601侵染后产生毛状根,其诱导率达36.6%;PCR结果表明,T-DNA序列已整合进橡胶树的基因组中。[结论]初步建立了发根农杆菌介导的橡胶树遗传转化体系。

【Abstract】 [Objective] To establish a new rubber tree genetic transformation system for the use of gene engineering technology to research rubber tree.[Method] The leaves and stems of rubber tree Reyan 7-33-97 were infected by Agrobacterium rhizogenes R1601,15834 and 1.2556 to induce the hairy roots,and then the PCR technique was used to detect the hairy roots induced by Agrobacterium rhizogenes.[Result] Under the same conditions,the hairy roots can be only successfully induced from rubber tree stems by Agrobacterium rhizogenes R1601,and its induction rate reached 36.6%;PCR results showed that T-DNA sequences have been integrated into the genome of rubber tree.[Conclusion] An Agrobacterium rhizogenes mediated rubber tree genetic transformation system has been preliminarily established.
