Lrh-1基因在湖羊HPG轴中表达量变化研究

【摘要】 [目的]揭示湖羊发情周期不同阶段下丘脑-垂体-性腺(HPG)轴组织中Lrh-1基因的表达变化规律。[方法]建立了湖羊Lrh-1基因的实时荧光定量(qRT-PCR)检测体系,测定其在发情前期、发情期、发情后期和间情期湖羊的下丘脑、垂体和卵巢间质组织中的表达量,分析Lrh-1基因在湖羊HPG轴的表达随发情周期变化的规律。[结果]建立的湖羊组织Lrh-1 mRNA检测体系扩增效率为92.44%,扩增效果良好。在湖羊的下丘脑、垂体和卵巢间质组织中均检测到Lrh-1基因mRNA的表达。其中,在发情后期下丘脑和卵巢间质组织中Lrh-1 mRNA相对表达量显著高于其他阶段的表达量(P<0.05),垂体组织中Lrh-1 mRNA相对表达量在发情周期不同阶段间差异不显著(P>0.05)。[结论]Lrh-1基因参与发情周期湖羊下丘脑、垂体和卵巢功能的行使,并且该基因的表达量变化可能与湖羊发情周期有关。

【Abstract】 [Objective] The aim of the study was to reveal the expression change laws of Lrh-1 gene in tissues of hypothalamus-pituitary-gonad(HPG) axis of Hu Sheep.[Method]The quantitative real-time PCR detection system(qRT-PCR) was established to detect the expression of Lrh-1 gene in Hu Sheep.The expression levels of Lrh-1 mRNA in tissues of hypothalamus,pituitary and gonad of Hu Sheep in proestrus,oestrum,diestrus and metoestrus were determined and the expression change laws of Lrh-1 gene in HPG axis of Hu Sheep were analyzed.[Result] The amplification efficiency of the established detection system of Lrh-1 in tissues of HU Sheep was 92.44% and the amplification effect was good.The expression of Lrh-1 mRNA was detected in the interstitial tissue of hypothalamus,pituitary and ovary of Hu Sheep.The relative expression level of Lrh-1 mRNA in the interstitial tissue of hypothalamus and ovary in metoestrus was significantly higher than that in other stages(P<0.05).The relative expression levels of Lrh-1 mRNA in pituitary tissues had no significant difference in different stages of estrous cycle(P>0.05).[Conclusion] Lrh-1 gene participated the function of hypothalamus,hypophysis,and ovary of Hu Sheep during estrous cycle,and the expression level changes of Lrh-1 might be related with the estrous cycle of Hu Sheep.
