不同花生品种胚小叶芽诱导的研究
Study on Shoot Induction from Leaflets of Different Peanut Cultivars(Arachis hypogaea L.)
【摘要】 比较了4个花生品种的胚小叶在不同浓度NAA和TDZ诱导培养基中的不定芽诱导率;并观察了用0.1%升汞和10%次氯酸钠不同处理的灭菌效果及其对种子萌发的影响。结果表明:花生种子用75%酒精处理0.5~1 min后用10%次氯酸钠处理15 min灭菌效果理想,萌发率高;诱导培养基中以添加1 mg/LNAA和0.02 mg/L TDZ最有利于不定芽诱导,4个花生品种诱导率都较高,达到80%以上,其中花育20的芽诱导率最高,为89.2%。
【Abstract】 The shoot induction rate of lobules embryos from 4 peanut cultivars were compared in the media added different concentrations of NAA and TDZ.The sterilizing effect of 0.1% HgCl2 and 10% NaClO and their influences to seed germination were also observed.The results indicated that the sterilizing effect was ideal and the germination rate was higher when the peanut seeds were treated for 0.5~1.0 minutes with alcohol and 15 minutes with 10% NaClO.The media added 1 mg/L NAA and 0.02 mg/LTDZ were the most favorable to in vitro shoot induction with the induction rate over 80%.In which,the shoot induction rate of Huayu 20 was the highest as 89.2%.
