清热化瘀方对脑缺血再灌注大鼠GDNF mRNA及其蛋白表达的影响

【摘要】 目的观察大鼠脑缺血再灌注后脑内胶质细胞源性神经营养因子(GDNF)mRNA和蛋白的表达变化以及清热化瘀方对其的影响。方法采用线栓法制作局灶性脑缺血大鼠(MCAO)模型,用原位杂交法和免疫组化法观察脑缺血后3h、6h、12h及1d、3d、7d共6个时间点GDNF mRNA及其蛋白的表达变化。结果缺血半暗带GDNF mRNA及其蛋白的表达均于缺血灌注后3h开始明显上升,6h达高峰,12h表达开始减弱(P<0.05或P<0.01),3d后降至正常水平。清热化瘀方治疗后可以上调GDNF的表达。结论清热化瘀方可促进脑缺血后GDNF的表达,保护神经元。

【Abstract】 Objective To observe the mRNA and protein levels of glial cell line-derived neurotrophic fator(GDNF) in rats after focal cerebral ischemia/reperfusion(I/R) and effect of Qingre Huayu prescription(QHP) on them.Methods The I/R rat models were established by middle cerebral artery occlusion to observe the expression variation of GDNF mRNA and protein at 3 h,6 h,12 h and 1 d,4 d,7 d after I/R by in situ hybridization and immunochemistry method.Results The expression of GDNF mRNA and protein in the penumbra region were increased at 3 h after reperfusion,and peaked at 6 hours,then decreased continuously at 12 h(P<0.05 or P<0.01).The level of GDNF was increased in QHP group.Conclusion Qingre Huayu prescription could protect neuron through upregulating the expression of GDNF after I/R in rats.
