酸樱桃汁对D-半乳糖致衰老小鼠抗氧化能力的影响

【摘要】 目的:观察酸樱桃汁对D-半乳糖致衰老模型小鼠抗氧化能力的影响。方法:将50只昆明小鼠随机分为正常对照组、模型组、酸樱桃汁低、中、高剂量组和维生素E组,除对照组外,其余5组分别颈背部皮下注射D-半乳糖120 mg/kg建立亚急性衰老模型;酸樱桃汁组分别灌胃酸樱桃汁(20,40,80g/kg),实验周期56d。治疗结束后,处死小鼠,取脾及胸腺称质量,计算脏器指数;分别测定小鼠脑组织中的脂褐素(LPF)含量,血清和肝脏中超氧化物岐化酶(SOD)活性,血清、肝脏和脑组织中的丙二醛(MDA)含量及谷胱甘肽过氧化物酶(GSH-Px)活性;结果:酸樱桃汁(40,80g/kg)能使小鼠胸腺和脾脏指数升高,脑组织中LPF水平下降,血清和肝脏中SOD水平提高;使血清、肝脏和脑组织中MDA含量下降,并使GSH-Px活力显著提高。结论:酸樱桃汁能显著提高亚急性衰老小鼠的抗氧化能力,具有一定的延缓衰老的作用。

【Abstract】 Objective:To investigate the effects of Prunus cerasus L juices(PCLJ) on antioxidative ability of senile mice induced by D-galactose.Methods:Fifty Kunming mice were randomly divided into 6 groups including control,model,vitmin E,and low dose of PCLJ(20g/kg·d) middle dose of PCLJ(40g/kg·d) and high dose of PCLJ(80g/kg·d),10 mice in each group.D-galactose(120mg/kg·d) was given to the mice in subcutaneous injection for 8 weeks successively to establish the mouse senile model.At the same time,PCLJ-treated mice were administered PCLJ intragastricly 20g/kg·d,40g/kg·d,and 80g/kg·d per day respectively.The indexes of spleen and thymus gland were determined.Lipofuscin level in brain,superoxide dismutase(SOD) activity in serum and live,and malondialdehyde(MDA) content and glutahione peroxidase(GSH-Px) activity in serum,live,and brain,were measured respectively to evaluate the antioxidatvie ability of PCLJ.Results:PCLJ administrations resulted in a significant increase in thymus index,spleen index of senile mice induced by D-galactose.PCLJ significantly decreased lipofusin levels in brain,and the levels of MDA in serum,liver,and brain.Moreover,PCLJ also increased SOD activity in serum and liver,and GSH-Px activity in serum,liver,and brain.Conclusion:PCLJ can effectively enhance the antioxidative ability of subacute senile mice induced by D-galactose.
