淡豆豉异黄酮对Ang Ⅱ诱导大鼠血管平滑肌细胞周期的影响

【摘要】 目的:观察淡豆豉异黄酮对血管紧张素Ⅱ(Angiotensin Ⅱ,Ang Ⅱ)诱导大鼠血管平滑肌细胞(Vascular Smooth MuscleCell,VSMC)细胞周期的影响,探讨其抑制增殖的可能机制。方法:原代培养大鼠VSMC,建立AngⅡ诱导VSMC增殖模型,MTT法观察淡豆豉异黄酮对Ang Ⅱ诱导VSMC增殖的干预作用,流式细胞仪观察淡豆豉异黄酮干预细胞后细胞周期的变化。结果:淡豆豉异黄酮50μg/L预孵育12、24 h,100μg/L预孵育1、4、12、24 h,200μg/L预孵育1、4、24 h,500μg/L预孵育4 h可显著抑制AngⅡ诱导的VSMC增殖;50、100、200μg/L作用24 h抑制增殖作用最为明显,并可显著增加VSMC G0/G1期比例,降低S期比例。结论:淡豆豉异黄酮抑制Ang Ⅱ诱导的VSMC增殖作用机制可能与阻止细胞由G0/G1期进入S期,进行VSMC细胞周期的调控有关。

【Abstract】 Objectives:To study the effect of semen sojae preparatum isoflavone(SSPI) on rat vascular smooth muscle cell(VSMC) cycle induced by Angiotensin Ⅱ(AngⅡ) and to explore the mechanism of inhibiting proliferation.Methods:VSMC of rat were primarily cultured,a cellproliferating model of VSMC induced by AngⅡ was established.The effect of SSPI on VSMC proliferation induced by Ang Ⅱ was analyzed by MTT method,the change of VSMC cell cycle was observed by flow cytometry.Results:SSPI of 50μg/L preincubated 12,24 h,100μg/L preincubated 1,4,12,24 h,200μg/L preincubated 1,4,24h,500μg/L preincubated 4h obviously inhibited the proliferation of VSMC induced by AngⅡ;SSPI of 50,100,200 μg/L that effected 24h most inhibited VSMC proliferation,the cell numbers in G0/G1 phase obviously increased and those of S phase decreased significantly.Conclusions:The proliferation of VSMC induced by AngⅡ can be inhibited by SSPI.The mechanism might be related to preventing VSMC from G0/G1 phase to S phase,and regulating VSMC cell cycle.
