芹菜素对大鼠心肌缺血再灌注损伤的保护作用
The protective effects of apigenin in myocardium of rats with ischemia reperfusion injury
【摘要】 目的:研究芹菜素对大鼠心肌缺血再灌注损伤的保护作用,并探讨其可能的作用机制。方法:采用结扎大鼠冠状动脉左前降支10min,再灌注10min,制成急性心肌缺血再灌注损伤模型。缺血前5 min舌下静脉推注芹菜素(1mg/kg、2mg/kg、4m/gkg),观察再灌10min后心肌组织丙二醛(MDA)的水平、超氧化物歧化酶(SOD)的活性和血清中乳酸脱氢酶(LDH)含量;用TTC染色法确定心肌损伤面积和梗死面积(MIS)。结果:芹菜素(4mg/kg)能降低再灌注心律失常总发生率和心律失常评分;芹菜素(2mg/kg、4mg/kg)显著减少缺血再灌注心肌MDA水平,增强SOD活性,剂量依赖性地降低LDH含量;芹菜素(1mg/kg、2mg/kg、4mg/kg)同时可剂量依赖性地缩小缺血再灌注大鼠的心肌梗死面积。结论:芹菜素对大鼠心肌缺血再灌注损伤具有保护作用。
【Abstract】 Objective: To study the protective effects of apigenin in myocardium of rats with ischemia reperfusion(I-R) injury and investigate the cardioprotective action problem mechanism.Methods: Myocardial ischemia reperfusion injury model was established through occluding the left anterior descending branch of coronary artery for 10min and removing the ligation later to reperfuse for 10min in vivo.We observed the incidence of arrhythmias and arrhythmia scores,we measured myocardial injury sizes and myocardial infarct sizes(MIS) by dying inury ventricles of I-R rat in TTC solution,and measured the level of serum latic dehydrogenase(LDH),myocardial superoside dismutase(SOD),myocardial malondialdehyde(MDA) through UV-Spectrometer.Compared with the I-R to explore its problem mechanism.Results : Apigenin(4 mg/kg) can suppress the incidence of arrhythmias,reduce arrhythmia scores(P<0.05);Apigenin(2mg/kg,4 mg/kg) can remarkably decreased the content of myocardial MDA(P<0.01);Apigenin(2mg/kg、4 mg/kg) can improved myocardial SOD activity(P<0.05);Apigenin(1mg/kg、2 mg/kg、4 mg/kg) can significantly reduce the level of serum LDH(P<0.01);Apigenin(1mg/kg、2 mg/kg、4 mg/kg) can shorten myocardial MIS(P<0.01).Conclusion: Apigenin can decrease the contents of myocardial MDA and serum LDH,shorten myocardial MIS,and increase myocardial SOD activity.Apigenin can prevent myocardial I-R arrhythmia.Therefore,apigenin plays a protective role on myocardial ischemia reperfusion injury.
