同型半胱氨酸对血管平滑肌细胞增殖周期的影响
The effect of Homocysteine on the cell cycle of VSMCs
【摘要】 目的研究表明,动脉粥样硬化(atherosclerosis,AS)的独立危险因子,同型半胱氨酸(homocysteine,Hcy),能通过促进血管平滑肌细胞(vascular smooth muscle cells,VSMCs)增殖而影响AS的发生,但Hcy与VSMCs增殖之间的关系,目前并不清楚。本研究采用不同浓度的Hcy刺激原代培养的脐静脉VSMCs,分析Hcy对VSMCs增殖周期的影响。方法常规培养人脐带VSMCs,将培养的VSMCs分为,0、50、100、200、500及1000μmol/L Hcy组,其中0μmol/LHcy组为对照组。当VSMCs细胞长满80%瓶底时,各组加入不同浓度的Hcy,继续培养细胞24h和48h,采用倒置显微镜观察细胞的生长状态,采用MTT法检测VSMCs的增殖活力,采用流式细胞技术分析VSMCs的增殖周期。结果当Hcy刺激VSMCs 24h和48h后,VSMCs的数目增多,细胞的活力增加(P<0.05);同时VSMCsS期细胞减少,G2/M期细胞增多,G0/G1细胞基本不变。结论 1.Hcy可直接诱导VSMCs的增殖;2.Hcy促进VSMCs从S期向G2/M期转变。
【Abstract】 Objective Homocysteine(Hcy),an independent risk foctor for atherosclerosis(AS),can induce proliferation of vascular smooth muscle cells(VSMCs),which is a key event in the genesis of the lesions of AS.However,the correlation between Hcy and proliferation of VSMCs is not clear.This study aims to investigate the impact of Hcy on the proliferation of VSMCs and the possible mechanism.Methods Cultured human umbilical VSMCs were divided 0,50,100,200,500 and 1000μmol/L Hcy groups,and the 0μmol/L Hcy group was the control.After the VSMCs were treated with different concentrations of Hcy for 24h and 48h,the morphology of cells was observed by inverted microscope,and the cell viability and the cell cycle were detected by MTT and flow cytometry(FCM) respectively.Results The number and viability of VSMCs was increased after the VSMCs were stimulated with Hcy for 24h and 48h(P<0.05),while the number of S phase of VSMCs was decreased and that of G2/M phase increased(P<0.05).Conclusions 1.Hcy could induce proliferation of VSMCs.2.Hcy could accelerate the conversion of the VSMCs of S phase into that of G2/M phase.
