HPLC-UV法测定大鼠血中姜黄素及一种新类似物的浓度
Determination of Curcumin and One of Its New Analogues in Rat Plasma by HPLC-UV
【摘要】 目的建立大鼠血浆中姜黄素及其一种新类似物T63的HPLC分析方法。方法含血浆样品以叔丁基甲醚进行液液萃取,取上清,HPLC进样分析。色谱条件:流动相为乙腈-水(60∶40),含5%冰醋酸。内标物:另一姜黄素新类似物T315。色谱柱:Hypersil BDS C18色谱柱(150 mm×4.6 mm,5μm)。紫外检测波长:355,415 nm。SD大鼠分别灌胃(28 mg.kg-1)与静注(5.6 mg.kg-1)姜黄素与T63后,以高效液相色谱法测定血药浓度。结果血浆中姜黄素的标准曲线范围为5.00 ng.mL-1~500.00 ng.mL-1,T63的标准曲线范围为10.0 ng.mL-1~500.0 ng.mL-1,回归方程分别为:Y=0.0049+0.0231X,r=0.9994;Y=-0.0115+0.0037X,r=0.9979;提取回收率分别为75.09%~83.12%,78.68%~86.70%(n=5);批内RSD分别≤10.20%、7.48%,批间RSD分别≤8.81%、7.41%。试验结果显示,T63的口服吸收较姜黄素有所改善。结论本分析方法灵敏、准确、重复性好,为姜黄素及其类似物在药代动力学研究中样品的检测提供了可靠的方法。
【Abstract】 Objective To establish a new,rapid and sensitive high performance liquid chromatography(HPLC) method to determine rat plasma curcumin and T63,one of the new analogues of curcumin.Methods After a single step liquid-liquid extraction with tert-butyl methyl ether,the extract supernatants were subjected to HPLC-UV anal-ysis.Chromatographic separation was performed on a BDS reversed-phase C 18 column(150 mm × 4.6 mm I.D.,parti-cle size 5 μm).The mobile phase consisted of acetonitrile and water(at the proportion of 60 ∶ 40),which contained 5 % acetic acid.T315,another new analogue of curcumin,was used for internal standard.The wave lengths for de-tection of curcumin and T63 were 415 nm and 355 nm,respectively.The concentration of curcumin and T63 in SD rat plasma was detected by using HPLC method after oral administration(28 mg·kg-1)and intravenous administration(5.6 mg·kg-1 respectively).Results The method had a chromatographic running time of 6 min,and had linear cali-bration curves at the concentrations of 5.0 ~ 500.0 ng/mL for curcumin and 10.0 ~ 500.0 ng/mL for T63.The regres-sion equations were Y=0.0049+0.0231X(r=0.9994)for curcumin and Y=-0.0115+0.0037X(r=0.9979)for T63 respec-tively.The recoveries of the method were 75.09 ~ 83.12 % for curcumin and 78.68 ~ 86.70 % for T63.The RSD of intra-batch precision was less than 10.20 % and 7.48 %,while the RSD of inter-batch precision was less than 8.81 % and 7.41 % for curcumin and T63,respectively.The validated method was successfully used to analyze the con-centrations of curcumin and T63 in rat plasma,and the results suggested that T63 has a better oral absorption than curcumin.Conclusion The method is accurate,reliable and practical,and is suitable for pharmacokinetic study of curcumin and its analogues in rats.
