清开灵颗粒对AD模型小鼠学习记忆能力的影响

【摘要】 目的研究清开灵颗粒对AD模型小鼠学习记忆的影响。方法选用东莨菪碱造模的雄性KM小鼠,用Morris水迷宫测试行为学指标,采用黄嘌呤氧化酶比色法测定超氧化物歧化酶(SOD)活性、硫代巴比妥酸法测定脂质过氧化产物丙二醛(MDA)含量、三硝基苯比色法测定脑内胆碱酯酶(AchE)活性。结果清开灵颗粒能明显缩短模型小鼠游泳时间及游泳路程;显著提高脑组织SOD活性,降低MDA含量;明显降低脑组织AchE活性。结论清开灵颗粒能改善造模小鼠的学习记忆功能,其机制可能与提高SOD活性、降低脑组织脂质过氧化物的含量、抑制AchE活性有关。

【Abstract】 Objective To study the effect of Qingkailing granule on learning and memory ability in Alzheimer’s dis-ease model mice.Methods Male KM mice were used for the experiment,and Alzheimer’s disease model was in-duced by scopolamine.Morris water maze was used to observe the learning and memory ability of model mice.Xan-thine oxidase colorimetry was used for the examination of superoxide dismutase(SOD)activity,thiobarbituric acid colorimetry for malondialdehyde content,and trinitrobenzene colorimetry for acetylcholinesterase(AchE)activity.Results Qingkailing granule could significantly shorten swimming time and swimming distance in Morris water maze,increase the activity of SOD,and decrease MDA content and AchE activity in the brain tissue of AD model mice.Conclusion Qingkailing granule could improve the ability of learning and memory of model mice,and its mechanism might involve in increasing SOD activity,reducing the level of peroxidation and inhibiting AchE activity in brain tis-sue.
