银杏外种皮提取物有效部位GBEE-2对胃癌SGC-7901细胞凋亡及其通路的影响

【摘要】 目的研究银杏外种皮提取物有效部位GBEE-2体外对人胃癌细胞凋亡及其凋亡通路的影响。方法体外培养人胃癌SGC-7901细胞,应用MTT法检测细胞增殖;应用流式细胞术检测凋亡率和细胞周期;应用ELISA法检测培养上清中基因蛋白Caspase-9、Fas及Survivin的含量。结果 GBEE-2(5~160μg.mL-1)对SGC-7901细胞的体外增殖具有抑制作用,并具有浓度依耐性,IC50为83μg.mL-1;在10~160μg.mL-1浓度下体外培养,GBEE-2可提高SGC-7901细胞的凋亡率,可使Caspase-9、Fas蛋白表达量增加(P<0.05或0.01),而Survivin蛋白的表达则减少(P<0.05或0.01)。结论 GBEE-2抑制人胃癌SGC-7901细胞的作用机制与诱导细胞凋亡有关,其凋亡通路涉及Caspase途径。

【Abstract】 Objective To study the influence of effective part GBEE-2 of Exocarpium Ginkgo extracts on apoptosis and apoptotic pathway of human gastric adenocarcinoma cell line SGC-7901.Methods SGC-7901 cells were cultured in vitro,and methyl thiazolyl tetrazolium(MTT) method was performed to observe the proliferation of SGC-7901.Flow cytometry was utilized for measuring the apoptotic rate and cell cycle.The expression of Caspase-9,Fas and Survivin in the cultured supernatant was detected by enzyme-labeled immunosorbent assay(ELISA).Results GBEE-2 could inhibit the proliferation of cell line SGC-7901 cells in vitro at the concentrations of 5~160 μg·mL-1,which was dose-dependent.IC50 of GBEE-2 was 83 μg·mL-1.When cultured in vitro with GBEE-2 at the concentrations of 10~160 μg·mL-1,the apoptotic rate of SGC-7901 was elevated,the expressions of Caspase-9 and Fas protein increased,but the expression of survivin protein decreased(P < 0.05 or P < 0.01).Conclusion GBEE-2 can inhibit the proliferation of SGC-7901 cells via the apoptosis induction,and caspase pathway is involved in the apoptosis pathway.
