【摘要】 目的:观察独角莲含药血清在体外对K562白血病细胞增殖及凋亡的影响。方法:采用血清药理学方法制备独角莲含药血清,以不同浓度的含药血清处理体外培养的K562白血病细胞,采用MTT比色法观察独角莲含药血清对K562细胞增殖的影响;采用流式细胞仪(FCM)及DNA片段凝胶电泳观察和检测细胞凋亡情况。结果:不同浓度独角莲含药血清对K562细胞增殖具有抑制作用,并呈剂量依赖关系。当含药血清作用96h后,其抑制作用开始减弱。流式细胞仪检测显示,独角莲含药血清能够诱导K562细胞的凋亡,且凋亡率随着剂量的增加而增大,与空白对照组比较(P<0.01);DNA琼脂糖凝胶电泳谱可见典型的DNA梯形带形成。结论:独角莲含药血清具有抑制K562白血病细胞增殖、促进细胞分化和诱导细胞凋亡的作用,其作用强度与时间-浓度呈正相关。独角莲含药血清对K562细胞发生细胞毒作用可能与诱导K562细胞凋亡有关。更多还原

【Abstract】 Objective: To study the effect of the herb serum containing HORN LIAN on the proliferation and apoptosis of K562 leukemia cells in vitro.Methods: Prepare the herb serum containing HORN LIAN with serologic pharmacological method and treat the K562 leukemia cells with contained herb serum of HORN LIAN in different concentrations.The effect of the herb serum on the proliferation of K562 cells was observed with MTT assay.Effect of the herb serum containing HORN LIAN on apoptosis of K562 cells was detected by flow cytometry and DNA agarose gel electrophoresis.Results: The herb-serum containing HORN LIAN in different concentrations could inhibit the proliferation of K562 leukemia cells directly in a dose-dependent.The inhibitive action began to attenuate after treatment with the drug-serum of HORN LIAN at 96h.The treated K562 cells by the herb serum containing HORN LIAN took on apoptosis by flow cytometry,and the herb serum containing HORN LIAN could increase the apoptosis rate of the K562 cells compared with blank control group with the increase of drug dose(P<0.01).The DNA ladder also was observed by agarose gel electrophoresis.Conclusion: The herb serum containing HORN LIAN could inhibit the proliferation,increase the differentiation and induce the apoptosis of K562 leukemia cells in a dose-time dependent manner.So we could conclude that cytotoxic action of the herb serum containing HORN LIAN may be related to the effect of inducing the apoptosis.更多还原
