活血祛瘀中药对大鼠矽肺模型的干预作用

【摘要】 目的研究活血祛瘀中药对SiO2所致矽肺模型肺组织中转化生长因子-β1(TGF-β1)、肿瘤坏死因子-α(TNF-α)及白介素-6(IL-6)表达的影响。方法将SD大鼠随机分为6组,分别为SiO2组、强的松+SiO2组、中药+SiO2组、强的松组、中药组、生理盐水组。末次给药后4 h,苯二胺盐酸盐法测定血清铜蓝蛋白(CP)含量;取肺组织,HE染色法行组织病理观察,免疫组化法测定肺组织肺组织胶原蛋白、TGF-β、TNF-α及IL-6的表达情况。结果治疗组与NS灌胃组病理检查未见明显差别;CP检测结果,SiO2+中药组较SiO2组、SiO2+西药组略有下降,但只与SiO2组比较差异有统计学意义;免疫组化法测定中药与对照组、西药治疗组肺组织胶原蛋白、TGF-β、TNF-α及IL-6的表达差异无统计学意义。结论在矽肺较严重的情况下,该抗矽肺方剂可能不能对矽肺模型起到干预的作用。

【Abstract】 [Objective]To study the effect of blood-activating and stasis-dissolving of Chinese medicine on the expression of TGF-β1,TNF-αand IL-6 in lung tissue of silicosis rats. [Methods]The SD Rats were randomly divided into 6 groups: SiO2,prednisone+SiO2,TCM+SiO2,prednisone,TCM,normal saline(NS).Four hours after last administration,serum ceruloplasmin(CP) was measured by phenylenediamine hydrochloride method;after HE staining of lung tissue,histopathologic examination was performed;the expression of collagen,TGF-β,TNF-α and IL-6 was determined by immunohistochemistry. [Results]No significant difference was found between treatment group and NS group by pathologic examination.The test results showed that CP contents was slightly reduced in TCM+SiO2 group,compared with SiO2 group and prednisone+SiO2 group,but compared with SiO2 group,the difference was significant.Immunohistochemistry determination showed that the expression of collagen,TGF-β,TNF-α and IL-6 in lung tissue was not significantly different between TCM group and control group as well as prednisone group. [Conclusion]This drug may not play the role of intervention on silicosis rats under the condition of serious silicosis.
