息风定颤汤对帕金森病模型大鼠多巴胺能神经元保护作用的研究

【摘要】 目的:观察息风定颤汤(以下简称中药)对PD模型大鼠旋转行为及对多巴胺能神经元的保护作用。方法:采用6-OHDA偏侧纹状体两点注入法制备PD模型大鼠,随机分为模型组、美多巴组、中药高、中、低剂量组,并设假手术组为对照组,灌胃治疗4周。观察各组大鼠治疗前后APO诱导的行为学变化及对纹状体单胺类神经递质、中脑TH的影响。结果:中药高、低剂量组灌胃后4周后旋转行为有明显改善(P<0.05,P<0.05);中药中剂量组灌胃2周、4周后旋转行为均有明显改善(P<0.05,P<0.01)。中药高、中、低剂量组大鼠损毁侧纹状体DA含量、HVA、DOPAC含量均明显增加,其中中剂量组增加最明显。中药高、中药、低剂量组TH阳性细胞表达均增加,与模型组相比,差异极显著(P<0.01,P<0.01,P<0.05),其中中药中剂量组增加最明显。结论:息风定颤汤可明显改善PD模型大鼠的旋转行为,增加纹状体内DA及其代谢产物的含量,上调TH的表达,对多巴胺能神经元具有保护作用,以中药中剂量组最明显。

【Abstract】 Objective: To observe the effect of Xifengdingchan Decoction on rotational behavior and neuroprotective effects on the Dopaminergie Neurons in Parkinson’s Disease Model Rats.Methods: PD model rats were made by injecting 6-OHDA two-point into unilateral striatum,randomly divided into model group,madopar group,herbal medicine high,medium and low dose group,sham-operated group established for the control group,treatment lasted for 4 weeks.The ethological change induced by apomorphine(APO),striatal monoaminergic neurotransmitter and TH in mesencephali of PD model rats were observed before treatment and 4 weeks’ treatment.Results:In high and low dose group improved obviously after 4 weeks’ treatment(P<0.05,P<0.05);in medium dose group improved obviously both after 2 and 4 weeks’ treatment(P<0.05,P<0.01).The content of DA,HVA,DOPAC in damaged lateral were all markedly increased in high,medium and low dose group;the medium dose group increased the most obviously.The expression of TH-positive cells were increased in high,medium and low dose group,Compared with model group,the difference was significant(P<0.01,P<0.01,P<0.05),in which medium group increased the most significantly.Conclusion : xifengdingchan tang could significantly improve the rotational behavior,increase the content of striatal DA and its metabolites and increase the expression of TH in PD rats,neuroprotective effects on the Dopaminergie Neurons in Parkinson’s Disease Model Rats,medium dose group was most obviously.
