补肾冲剂联合干扰素治疗慢性乙型肝炎及其对树突状细胞的影响
Clinical Effect and Affect on Dentritic Cells Function of Bushen Recipe Combined with Interferon on CHB Patients
【摘要】 目的:观察干扰素联合补肾冲剂治疗慢性乙型肝炎(chron ic hepatitis B,CHB)的疗效,探讨该药物对CHB患者树突状细胞(dentritic cells,DCs)的影响。方法:收集CHB患者60例,随机分为治疗组及对照组,治疗组给予补肾冲剂联合干扰素治疗,对照组单用干扰素治疗,疗程均为6个月。分离扩增外周血单个核细胞中DC,流式细胞法检测DC表面CD86、CD80、CD40、CD14、CD11c、HLA-DR的表达。检测同种异体T淋巴细胞混合培养后的刺激指数及培养上清液中IFN-γ、IL-10表达。结果:治疗后治疗组ALT、AST改善较对照组明显,P<0.05;治疗后治疗组HBVDNA对数均值明显低于对照组,具有显著差异(P<0.05)。治疗后治疗组淋巴细胞混合实验中IFN-γ明显高于对照组(P<0.05)。结论:中药补肾冲剂与干扰素联合治疗CHB在改善肝功能、降低HBVDNA数量方面疗效更,且对IFN-γ有明显的升高作用,提示其疗效机理可能在于提高患者的细胞免疫能力。
【Abstract】 Objective:To observe the effect of Bushen recipe combined with interferon on chronic hepatitis B(CHB) and study the effect on dentritic cells(DCs) function in CHB patients.Methods:A total of 60 CHB patients were enrolled and randomly divided into treatment and control groups,patients in the treatment group were given Bushen recipe combined with interferon therapy,while patients in the control group were only given interferon therapy.Both the therapies lasted for 6 months.DCs were isolated from peripheral blood and cultured,then the expression of surface markers on DCs(CD86,CD80,CD40,CD14,CD11c and HLA-DR) were detected before and after treatment by flow cytometry.The stimulation index of allogeneic T cells after co-culture was detected,as well as the expression of IFN-γ and IL-10 in co-culture supernatants.Results:The level of ALT,AST and HBV DNA significantly decreased in treatment group compared to control group after therapy(P<0.05).The expression of IFN-γ in allogeneic lymphocytes co-cultured assay significantly increased in treatment group compared to control group after therapy(P<0.05).Conclusion:The combination therapy of Bushen recipe and interferon showed good effect on the improvement of liver function,the reduction of viral load and the increase of IFN-γ.The mechanism may relate to the improvement of cell immunocompetent.
