【摘要】 目的:研究新型降血糖化合物G004对自发性糖尿病kkay小鼠糖脂代谢的影响。方法:采用自发性2型糖尿病kkay小鼠连续灌胃给药,测定血糖、血脂和血糖调节激素水平;采用β-TC-6胰岛素分泌细胞,测定G004对细胞分泌胰岛素的作用;制备大鼠体外灌流胰腺,放射免疫法测定灌流流出液中的胰岛素含量。结果:G004有效降低kkay小鼠空腹血糖、三酰甘油水平,并显著降低游离脂肪酸;G004促进β-TC-6细胞及体外灌流胰腺分泌胰岛素。结论:化合物G004可能通过促进胰岛素分泌改善kkay小鼠糖代谢和脂代谢。更多还原

【Abstract】 Objective: To investigate the effect of G004 on glucose and lipid metabolism in type 2 diabetic kkay mice.Methods: The effect of G004 on blood glucose,lipid and the hormones regulating blood glucose was examined in type 2 diabetic kkay mice.The insulin-secreting cell line β-TC-6 cells were used to examine its effect on stimulation of insulin secretion.Isolated rat pancreas was isolated and perfused with G004,and insulin concentrations in effluent perfusate were measured by radioimmunoassay(RIA).Results: The fasting plasma glucose,triglyceride level and free fatty acids levels were effectively reduced in kkay mice treated with G004.Insulin secretion was effectively promoted by G004 from the perfused pancreas and β-TC-6 cells.Conclusion: G004 stimulates insulin secretion and ameliorates the abnormity of glucose and lipid metabolism in kkay mice.更多还原
