靛玉红对溃疡性结肠炎小鼠肠黏膜细胞因子含量的影响

【摘要】 [目的]探讨中药青黛有效成分靛玉红对溃疡性结肠炎(UC)小鼠肠黏膜细胞因子含量的影响。[方法]采用5%葡聚糖硫酸钠(DSS)建立UC小鼠模型,将小鼠随机分为靛玉红高、低剂量组,美沙拉嗪组,模型1、2组和正常组,每组10只。其中模型组于造模7d时处死,其余各组于第7d开始灌胃给药,连续给药7d时处死。ELISA法检测结肠组织中肿瘤坏死因子α(TNF-α)和白细胞介素4(IL-4)含量。[结果]与正常组比较,模型1组结肠黏膜TNF-α含量增高,IL-4含量减少,差异有统计学意义(P<0.05);与模型2组比较,靛玉红高、低剂量组和美沙拉嗪组TNF-α含量减少,IL-4含量增加,差异有统计学意义(P<0.05)。[结论]靛玉红减少TNF-α含量、增加IL-4含量可能是其治疗UC的主要作用机制。

【Abstract】 [Objective]To investigate the effect of indirubin on cytokines in colonic mucosa of dextran sulfate sodium(DSS)-induced colitis in mice.[Methods]Mouse colitis model was induced by oral administration of 5% DSS for 7 days,and 7-day intervention treatment was carried out after modeling.Mice were divided into normal control group,colitis model group 1,colitis model group 2,low-dose indirubin group,high-dose indirubin group and 5-aminosalicylic acid(5-ASA)group.After the treatment,IL-4 and TNF-α levels in the colonic tissue were detected with ELISA.[Results]Colonic IL-4 level significantly decreased in colitis model group 1 while the TNF-α level significantly increased compared with normal control group(both P<0.05).Colonic IL-4 level increased in all the treatment groups while the TNF-α level decreased compared with colitis model group 2(both P<0.05).[Conclusion]The major mechanism of indirubin’ effect on ulcerative colitis may be due to increased IL-4 level and decreased TNF-α level.
